      Abstract
(>245 kDa)

Collagen is a functional biomaterial with high economic value and has been widely utilized across various industrial sectors. Sailfish (Istiophorus sp.) skin waste has the potential to be utilized as an alternative collagen source to reduce import dependency while increasing the added value of fishery by-products. This study aimed to optimize extraction conditions and characterize Bromelain Soluble Collagen (BSC) from sailfish skin by varying pretreatment conditions and extraction temperature. Pretreatment using NaOH (0.1–0.6 M) with ultrasonic assistance was followed by extraction with 0.5 M CH3COOH and bromelain at 4 °C and 30 °C. Optimal treatment (0.1 M NaOH, 15 h, 4 °C) produced a collagen yield of 12.04 ± 0.34%. The resulting BSC exhibited a pH of 4.38 ± 0.37, whiteness degree of 82.65 ± 0.41, Hyp/Protein ratio of 0.295, imino acid content of 0.624, and collagen conversion factor of 3.39. Electrophoretic analysis revealed the a1 (130 kDa), a2 (100 kDa), ß (245 kDa), and ? (>245 kDa) polypeptide chains, while FTIR analysis showed the characteristic amide A, B, I, II, and III bands.
