Karakterisasi Gen Ghrelin (GHRL) pada Ayam Kampung Ayam Leher Gundul Ayam Walik dan Ayam Arab
Abstract
Ghrelin gene (GHRL) is one of important gene that stimulates secretion of gastric acid, growth hormone and food intake. Sekuens of GHRL fragment in this study was located in promoter, exon 1 and partial intron 1. This study aimed to characterize GHRL gene especially to detect mutation sites of kampong, naked neck, walik, and arab chicken. Twenty samples were amplified to result of fragment with length of 1 050 bp. Characterization of GHRL gene was processed by BLAST-N, then MEGA 5 was used to analize the sequence data. Annealing temperature was performed at 60 ºC for 1 minute. The alignment product of eight samples by using primer forward produced sequence of 554 bp. The similarity value of GHRL gene fragment with sequence data on GenBank (AB158617) was 99%. There was found 3 mutation sites of GHRL gene fragment. However, the 554 bp of GHRL gene fragment has low power to distinguish chicken breeds since all the samples have pairwise distance based on the phylogenetic tree construction analysis.
