| dc.description.abstract | Buffered Peptone Water (BPW) merupakan media pengencer yang penting dalam mempertahankan viabilitas mikroorganisme sebelum pengujian Total Plate Count (TPC). Penelitian ini mengevaluasi pengaruh suhu penyimpanan (dingin 2–8°C dan ruang 25–28°C) dan jenis penutupan (sponge plug dengan dan tanpa aluminium foil) terhadap nilai Angka Lempeng Total (ALT) dan pH larutan BPW selama tujuh hari penyimpanan pada sampel tepung bumbu serbaguna. Rancangan Acak Lengkap digunakan dengan 28 kombinasi perlakuan dan satu kontrol, masing-masing tiga ulangan, dianalisis dengan One-Way ANOVA dan uji lanjut Dunnett (a=0,05). Hasil menunjukkan kombinasi hari dan perlakuan berpengaruh sangat nyata (P<0,05) terhadap ALT dan pH. Penyimpanan suhu dingin dengan aluminium
foil (F1) paling efektif mempertahankan mutu BPW hingga hari ke-7, sedangkan suhu ruang tanpa aluminium foil (F4) mengalami penurunan ALT terbesar pada hari ke-7, sejalan dengan penurunan pH dengan pola F1<F2<F3<F4. BPW direkomendasikan disimpan pada suhu dingin dengan sponge plug dan aluminium foil, maksimal 7 hari, untuk menjaga akurasi pengujian mikrobiologi di PT XYZ. | |
| dc.description.abstract | Buffered Peptone Water (BPW) is a diluent medium important for maintaining microbial viability prior to Total Plate Count (TPC) testing. This study evaluated the effect of storage temperature (cold 2–8°C and room 25–28°C) and closure type (sponge plug with and without aluminium foil) on the TPC and pH values of BPW solution over seven days of storage, using multipurpose seasoning flour as the sample. A Completely Randomized Design with 28 treatment combinations and one control, each replicated three times, was analyzed using One-Way ANOVA and Dunnett's test (a=0.05). Results showed that storage duration and treatment type had a highly significant effect (P<0.05) on TPC and pH. Cold storage with aluminium foil (F1) was most effective in maintaining BPW quality until day 7, whereas room temperature storage without aluminium foil (F4) showed the largest TPC decline by day 7, consistent with a pH decline pattern of F1<F2<F3<F4. BPW is recommended to be stored at cold temperature with sponge plug and aluminium foil, for a maximum of 7 days, to maintain the accuracy of microbiological testing at PT XYZ. | |
