Penelitian ini bertujuan mengembangkan metode screening cepat mutu mikrobiologi susu segar berbasis Color Detector pada uji resazurin di PT XYZ. Penelitian dilakukan dalam tiga tahap, yaitu verifikasi prosedur spiking bakteri dan optimasi metode resazurin, penyusunan skala mutu, serta evaluasi kinerja menggunakan enam sampel independen. Hasil menunjukkan prosedur spiking menghasilkan tingkat cemaran sesuai target dengan bias log < 0,5 log CFU/mL. Hue ditetapkan sebagai parameter utama dengan korelasi kuat terhadap jumlah mikroorganisme (R = 0,94), sedangkan Red sebagai parameter pendukung (R = 0,70) dengan waktu inkubasi optimum 30 menit. Nilai Hue menunjukkan perbedaan signifikan antara Staphylococcus aureus dan Escherichia coli (p = 0,045). Skala mutu terdiri atas kategori sangat baik, baik, dan jelek. Evaluasi menghasilkan akurasi 83,33%, sensitivitas 100%, dan spesifisitas 80%, sedangkan Fisher’s exact test menghasilkan p = 0,050. Metode berpotensi digunakan sebagai screening awal dengan waktu analisis 30 menit dan telah diimplementasikan melalui dashboard, decision tree (pohon keputusan), dan Instruksi Kerja (IK). This study aimed to develop a rapid microbiological quality screening method for fresh milk based on the Color Detector application and resazurin test at PT XYZ. The research was conducted in three stages: verification of the bacterial spiking procedure and optimization of the resazurin method, development of a quality scale, and performance evaluation using six independent samples. The results showed that the spiking procedure produced contamination levels within the target range, with a log bias of < 0,5 log CFU/mL. Hue was selected as the primary parameter due to its strong correlation with the number of microorganisms (R = 0,94), while Red was used as a supporting parameter (R = 0,70), with an optimum incubation time of 30 minutes. Hue values showed a significant difference between Staphylococcus aureus and Escherichia coli (p = 0,045). The quality scale consisted of three categories: very good, good, and poor. Performance evaluation resulted in an accuracy of 83,33%, sensitivity of 100%, and specificity of 80%, while Fisher’s exact test yielded p = 0,050. The method has potential as a preliminary screening method with a 30-minute analysis time and has been implemented through a dashboard and decision tree.
