      Desain Bioaugmentasi Menggunakan Bio-OSD dan Sel Vegetatif Bacillus pseudomycoides RAY21 pada Bioremediasi Tanah Tercemar Minyak Bumi

      Abstract
      Pencemaran tanah oleh minyak bumi merupakan permasalahan lingkungan yang memerlukan teknologi remediasi yang efektif dan ramah lingkungan. Penelitian ini bertujuan menghasilkan desain bioaugmentasi berbasis Bio-Oil Spill Dispersant (Bio-OSD) dan sel vegetatif Bacillus pseudomycoides RAY21 untuk meningkatkan efektivitas bioremediasi tanah tercemar minyak bumi. Pengembangan desain dilakukan melalui simulasi bioremediasi menggunakan tanah dengan kadar awal Total Petroleum Hydrocarbon (TPH) sebesar 2% dan 0,5% sebagai representasi tingkat pencemaran yang berbeda. Strategi yang dikembangkan diawali dengan aplikasi Bio-OSD untuk meningkatkan bioavailabilitas hidrokarbon, kemudian dilanjutkan dengan inokulasi sel vegetatif B. pseudomycoides RAY21 serta evaluasi melalui dua iterasi pengujian. Parameter yang diamati meliputi TPH, viabilitas bakteri, kadar air, pH, dan suhu. Hasil menunjukkan bahwa kombinasi Bio-OSD dan sel vegetatif B. pseudomycoides RAY21 mampu menurunkan kadar TPH pada kedua tingkat pencemaran. Evaluasi iterasi pertama menunjukkan penurunan viabilitas bakteri pada akhir inkubasi sehingga dilakukan penyempurnaan desain melalui augmentasi ulang (re-augmentation) menggunakan sel vegetatif. Desain bioaugmentasi yang dikembangkan memberikan kinerja terbaik pada tanah dengan TPH awal 0,5% yang mencapai kadar TPH akhir sebesar 0,12%, sedangkan pada tanah dengan TPH awal 2% masih diperlukan optimasi untuk meningkatkan efisiensi biodegradasi. Desain yang dikembangkan menunjukkan potensi untuk meningkatkan efektivitas bioremediasi melalui strategi augmentasi bertahap (staged bioaugmentation) serta menjadi dasar implementasi proses bioremediasi pada skala lapangan.
       
      Petroleum-contaminated soil is an environmental issue that requires effective and environmentally friendly remediation technologies. This study aimed to develop a bioaugmentation design employing Bio-Oil Spill Dispersant (Bio-OSD) and vegetative cells of Bacillus pseudomycoides RAY21 for the bioremediation of petroleum-contaminated soil. The bioaugmentation design was developed through laboratory-scale bioremediation simulations using soils with initial Total Petroleum Hydrocarbon (TPH) concentrations of 2% and 0.5%, representing different contamination levels. The proposed strategy consisted of an initial Bio-OSD application to enhance hydrocarbon bioavailability, followed by inoculation with vegetative cells of B. pseudomycoides RAY21 and evaluation through two experimental iterations. The observed parameters included TPH, bacterial viability, moisture content, pH, and temperature. The results demonstrated that the combination of Bio-OSD and vegetative cells of B. pseudomycoides RAY21 reduced TPH concentrations at both contamination levels. Evaluation of the first iteration revealed a decline in bacterial viability at the end of the incubation period, leading to refinement of the design through vegetative cell re-augmentation. The developed bioaugmentation design achieved its best performance in soil with an initial TPH concentration of 0.5%, resulting in a final TPH concentration of 0.12%, whereas further optimization was required for soil with an initial TPH concentration of 2% to improve biodegradation efficiency. Overall, the developed design demonstrates the potential to enhance bioremediation performance through a staged bioaugmentation strategy and provides a basis for future implementation of petroleum-contaminated soil bioremediation at the field scale.
