dc.description.abstractProyek ini dilatarbelakangi oleh perlunya teknologi bioremediasi yang efektif untuk menangani tanah tercemar minyak bumi yang sulit dipulihkan karena minyak terjerap kuat pada matriks tanah. Tujuan penelitian ini adalah mengevaluasi efektivitas Bio-OSD pada berbagai rasio Dispersant to Oil Ratio (DOR) dalam mendispersikan dan menurunkan total petroleum hydrocarbon (TPH) pada tanah tercemar minyak, menentukan DOR optimum, serta menilai kinerja Bio-OSD yang dikombinasikan dengan Bacillus pseudomycoides RAY21 dalam bentuk endospora dan sel vegetatif. Metode penelitian meliputi pengujian mikroskopis, analisis TPH. Hasil menunjukkan bahwa DOR 0,5:1 merupakan perlakuan terbaik karena memberikan rata-rata penurunan TPH paling tinggi dibandingkan variasi DOR lainnya. Pada aplikasi dengan B. pseudomycoides RAY21, formulasi endospora menunjukkan dispersi minyak lebih stabil dan penurunan TPH lebih besar dibandingkan sel vegetatif, dengan capaian penurunan tertinggi pada tanah tercemar 0,5% sebesar 95,3% dan pada tanah tercemar 2% sebesar 81,0%. Temuan ini menunjukkan bahwa kombinasi Bio-OSD dan endospora Bacillus berpotensi meningkatkan bioavailabilitas hidrokarbon dan efisiensi bioremediasi tanah tercemar minyak bumi.
dc.description.abstractThis project is motivated by the need for effective bioremediation technologies to restore petroleum-contaminated soils that are difficult to remediate due to strong oil adsorption onto the soil matrix. The objectives of this study were to evaluate the effectiveness of Bio-OSD at various Dispersant to Oil Ratios (DOR) in dispersing and reducing total petroleum hydrocarbons (TPH) in contaminated soil, determine the optimum DOR, and assess the performance of Bio-OSD combined with Bacillus pseudomycoides RAY21 in both endospore and vegetative cell forms. Research methods included microscopic analysis and TPH determination. Results showed that DOR 0.5:1 was the most effective treatment, achieving the highest average TPH reduction compared to other DOR variations. In applications with B. pseudomycoides RAY21, the endospore formulation demonstrated more stable oil dispersion and greater TPH reduction than vegetative cells, with the highest reductions of 95.3% in 0.5% contaminated soil and 81.0% in 2% contaminated soil. These findings indicate that combining Bio-OSD with Bacillus endospores has the potential to enhance hydrocarbon bioavailability and improve the efficiency of petroleum-contaminated soil bioremediation.
