      Abstract
      Whey merupakan hasil samping industri susu yang masih mengandung nutrien dan berpotensi dimanfaatkan sebagai media pertumbuhan mikroorganisme. Penelitian ini bertujuan mengevaluasi potensi whey sebagai media alternatif untuk pertumbuhan Bacillus amyloliquefaciens SLBD serta menentukan konsentrasi whey yang optimal. Kombinasi medium whey dan (Luria Bertani/LB) menggunakan konsentrasi whey yang berbeda (0%, 25%, 50%, 75%, dan 100%). Pertumbuhan bakteri dianalisis menggunakan optical density (OD), total plate count (TPC), dan SDS-PAGE. Hasil menunjukkan bahwa whey memiliki karakteristik sweet whey yang mendukung pertumbuhan bakteri. Konsentrasi whey 50% memberikan hasil terbaik dengan jumlah bakteri hidup mencapai 5,38 Log CFU mL-1. Analisis SDSPAGE menunjukkan bahwa suplementasi whey 25% – 50% mampu menginduksi ekspresi protein adaptif tambahan. Temuan ini menunjukkan bahwa whey 50% berpotensi digunakan sebagai media kultur alternatif yang ekonomis dan ramah lingkungan untuk kultivasi B.amyloliquefaciens SLBD.
       
      Whey is a dairy industry by-product rich in nutrients and has potential as a microbial growth medium. This study aimed to evaluate whey as an alternative medium for the growth of Bacillus amyloliquefaciens SLBD and to determine the optimal whey concentration. A combination of whey and LB media using different concentrations of whey (0%, 25%, 50%, 75%, and 100%). Bacterial growth was assessed using optical density (OD), total plate count (TPC), and SDS-PAGE analysis. The results showed that the whey exhibited sweet whey characteristics that supported bacterial growth. The 50% whey treatment produced the best performance, with a viable cell count of 5,38 Log CFU mL-1. SDS-PAGE analysis revealed that 25% – 50% whey supplementation induced the expression of additional adaptive proteins. These findings indicate that 50% whey has potential as an economical and environmentally friendly alternative culture medium for B. amyloliquefaciens SLBD.
