      Karakterisasi Dua Produk Ulvan Hasil Presipitasi Isopropil Alkohol pada Ekstraksi Ultrasonikasi dengan pH Pelarut

      Abstract
      Ulvan merupakan polisakarida tersulfatasi yang terdapat pada dinding sel rumput laut hijau Ulva sp. dan berpotensi dimanfaatkan pada bidang pangan, biomaterial, dan biomedis. Penelitian ini bertujuan menganalisis pengaruh variasi pH pelarut terhadap karakteristik fisiko-kimia ulvan hasil ekstraksi berbantu ultrasonikasi serta menentukan perlakuan terbaik. Ekstraksi dilakukan menggunakan pelarut dengan pH 5, 7, dan 9. Hasil penelitian menunjukkan bahwa variasi pH pelarut berpengaruh nyata terhadap bobot ampas, kadar air, kadar abu, viskositas, dan bobot molekul ulvan. Perlakuan pH 9 menghasilkan rendemen tertinggi sebesar 18,45% dan bobot ampas terendah sebesar 60,95%, sehingga dipilih sebagai perlakuan terbaik. Karakterisasi ulvan terpilih menunjukkan keberadaan gugus hidroksil, karboksilat, dan sulfat sebagai gugus fungsi khas ulvan. Fraksi bawah memiliki kadar sulfat dan kadar protein yang lebih tinggi dibandingkan fraksi atas, yaitu masing-masing sebesar 19,61% dan 10,18%. Hasil penelitian menunjukkan bahwa pH pelarut berpengaruh terhadap karakteristik ulvan yang dihasilkan melalui ekstraksi berbantu ultrasonikasi.
       
      Ulvan is a sulfated polysaccharide found in the cell wall of the green seaweed Ulva sp. and has potential applications in the food, biomaterial, and biomedical sectors. This study aimed to evaluate the effect of solvent pH variation on the physicochemical characteristics of ulvan extracted using ultrasonic-assisted extraction and to determine the optimum extraction condition. Ulvan was extracted using solvents adjusted to pH 5, 7, and 9. The results showed that solvent pH significantly affected the residue weight, moisture content, ash content, viscosity, and molecular weight of ulvan. The pH 9 treatment produced the highest yield (18.45%) and the lowest residue weight (60.95%) and was therefore selected as the best treatment. Characterization of the selected ulvan revealed the presence of hydroxyl, carboxylate, and sulfate groups. The bottom fraction exhibited higher sulfate and protein contents than the upper fraction, reaching 19.61% and 10.18%, respectively. These findings indicate that solvent pH influences the characteristics of ulvan obtained through ultrasonic-assisted extraction.
