Perkembangan Embrio Samia cynthia ricini dengan Variasi Lama Perendaman Metanol 2 M pada Penyimpanan Suhu Rendah
Abstract
Penyesuaian waktu antara peternak inti dan peternak plasma menjadi tantangan dalam budidaya ulat sutra eri (Samia cynthia ricini). Penelitian ini bertujuan menganalisis pengaruh lama perendaman metanol 2 M dan lama penyimpanan suhu rendah dalam inkubator terhadap perkembangan embrio, durasi penetasan, penundaan penetasan, dan daya tetas telur S. c. ricini. Penelitian menggunakan rancangan acak lengkap faktorial dengan dua faktor, yaitu lama perendaman metanol (3, 5, dan 7 menit) dan lama penyimpanan inkubator (7 dan 10 hari) dengan tiga ulangan. Data dianalisis menggunakan ANOVA dua arah dan uji Tukey. Hasil penelitian menunjukkan bahwa penyimpanan suhu rendah memperlambat perkembangan embrio. Penyimpanan selama 7 dan 10 hari menghasilkan penundaan penetasan masing-masing selama 10 dan 13 hari. Lama penyimpanan inkubator berpengaruh sangat nyata sedangkan lama perendaman metanol tidak berpengaruh nyata. Interaksi kedua faktor hanya berpengaruh nyata terhadap durasi penetasan dan penundaan penetasan. Perendaman metanol selama 5 menit dengan penyimpanan inkubator 7 hari menghasilkan daya tetas sekitar 26%. Synchronizing the production schedules between nucleus and plasma farmers remains a challenge in eri silkworm (Samia cynthia ricini) cultivation. This study aimed to evaluate the effects of 2 M methanol immersion duration and low-temperature storage period in an incubator on embryonic development, hatching duration, hatching delay, and hatchability of S. c. ricini eggs. A completely randomized factorial design with two factors was employed: methanol immersion duration (3, 5, and 7 min) and incubator storage period (7 and 10 days), with three replications. Data were analyzed using two-way analysis of variance (ANOVA) followed by Tukey's post hoc test. The results showed that low-temperature storage slowed embryonic development. Storage for 7 and 10 days delayed hatching by 10 and 13 days, respectively. The incubator storage period had a highly significant effect, whereas methanol immersion duration had no significant effect. A significant interaction between the two factors was observed only for hatching duration and hatching delay. The combination of 5-min methanol immersion and 7-day incubator storage resulted in a hatchability of approximately 26%.
