dc.description.abstractKerbau memiliki sistem pencernaan yang kompleks dan kaya akan komunitas mikroorganisme. Penelitian ini bertujuan mengarakterisasi komposisi serat, profil mikrobiota, serta DNA genomik pada feses kerbau Toraja segar dan tidak segar yang berpotensi penghasil enzim hidrolitik. Sampel feses dikoleksi dari peternakan tradisional di Enrekang, Sulawesi Selatan, dan dianalisis melalui pengujian TPC, BAL, aktivitas selulolitik, analisis komposisi serat menggunakan metode Van Soest, serta ekstraksi DNA genomik menggunakan Kit PrimeWay Genomic II. Data dianalisis menggunakan uji t independen (P < 0,05). Hasil menunjukkan bahwa feses segar memiliki nilai TPC sebesar 7,93 Log CFU g-1, BAL sebesar 5,37 Log CFU g-1, dan IS sebesar 2,09 yang nyata lebih tinggi dibandingkan dengan feses tidak segar, disertai kandungan NDF dan hemiselulosa yang lebih tinggi. Homogenisasi mekanik dapat meningkatkan konsentrasi DNA pada feses segar, namun kemurnian DNA masih di bawah standar sehingga optimasi ekstraksi masih diperlukan. Feses segar kerbau Toraja terbukti menjadi sumber potensial bakteri indigenous penghasil enzim hidrolitik untuk pengembangan bioteknologi berbasis sumber daya lokal.
dc.description.abstractBuffalo possess a complex digestive system richly populated by diverse microbial communities. This study aimed to characterize the fiber composition, microbial profile, as well as DNA in fresh and non-fresh feces of Toraja buffalo as potential producers of hydrolytic enzymes.. Fecal samples were collected from traditional livestock farms in Enrekang, South Sulawesi, and analyzed through TPC, LAB, cellulolytic activity screening, fiber composition analysis using the Van Soest method, and genomic DNA extraction using the PrimeWay Genomic II Kit. Data were analyzed using an independent samples t-test (P < 0.05). Results showed that fresh feces yielded significantly higher TPC 7,93 Log CFU g-1, LAB count 5,37 Log CFU g-1, and CI 2,09 compared to unfresh feces, accompanied by higher NDF and hemicellulose content. Mechanical homogenization can increase DNA concentration in fresh feces; however, DNA purity remained below the standard, indicating that further extraction optimization is still required. Fresh feces of Toraja Buffalo were proven to be a potential source of indigenous bacteria producing hydrolytic enzymes for local resource-based biotechnology development.
