      Pengaruh Penambahan Biostimulan Berbahan Baku Kulit Nipah Terhadap Stabilitas Bakteri Denitrifikasi Di Media Budidaya Udang Vaname

      Abstract
      Bakteri denitrifikasi memiliki kemampuan dalam mengkonversi limbah kegiatan budidaya berupa nitrogen anorganik berbahaya menjadi gas nitrogen yang dilepaskan ke atmosfer. Penambahan tepung kulit nipah berpotensi menjadi sumber karbon organik yang mendukung pertumbuhan bakteri denitrifikasi. Penelitian ini bertujuan menentukan dosis terbaik penambahan tepung kulit buah nipah terhadap stabilitas bakteri denitrifikasi dan parameter kualitas air pemeliharaan udang vaname. Perlakuan yang diberikan terdiri dari kontrol, 20 mg L-1, 40 mg L-1, dan 60 mg L-1 dengan tiga kali pengulangan pada setiap perlakuan. Pemberian tepung kulit nipah hanya diberikan satu kali sebelum udang uji masuk ke dalam wadah pemeliharaan. Udang uji yang digunakan adalah udang vaname stadia PL 25. Pemeliharaan udang uji dilakukan selama 30 hari tanpa pergantian air menggunakan wadah boks kontainer berukuran 30 L. Perhitungan kelimpahan bakteri denitrifikasi dilakukan satu kali pada hari ke-30 pemeliharaan. Hasil pemeliharaan udang vaname dengan penambahan tepung kulit nipah 60 mg L-1 memberikan hasil terbaik terhadap kelimpahan bakteri denitrifikasi dengan jumlah mencapai 3,3 x 106 CFU mL-1. Hasil identifikasi bakteri menunjukkan bahwa bakteri yang ditemukan diduga tergolong ke dalam bakteri Acinetobacter sp. Kelimpahan bakteri denitrifikasi yang dihasilkan pada perlakuan dosis 60 mg L-1 mampu menekan kadar nitrat hingga 0,73±0,03 mg L-1 di hari ke-30 pemeliharaan. Penambahan tepung kulit nipah dengan dosis 60 mg L-1 merupakan dosis paling efektif dalam meningkatkan kelimpahan bakteri denitrifikasi dan menjaga kondisi lingkungan budidaya.
       
      Denitrifying bacteria are crucial to convert cultivation waste in the form of harmful inorganic nitrogen into nitrogen gas that is released to the atmosphere. The addition of nipa fruit skin flour has the potential as a source of organic carbon that supports the growth of denitrifying bacteria. This study investigated the potential of nipa fruit skin flour as an organic carbon source to enhance denitrifying bacteria in shrimp cultivation. The treatments given consisted of control, 20 mg L-1, 40 mg L-1, and 60 mg L-1 with three repetitions in each treatment. Nipa fruit skin flour was only given once before the shrimp entered the container. The shrimp used at the stage PL 25. The shrimp were maintained in 30L container box for 30 days without water changes. The calculation of the abundance of denitrifying bacteria was conducted on the 30th day of cultivation. The results of the addition of 60 mg L-1 nipa fruit skin flour produced an abundance of denitrifying bacteria with a number reaching 3,3 x 106 CFU mL-1. The results of bacterial identification indicate that the bacteria found are classified as Acinetobacter sp. The abundance of denitrifying bacteria produced in the 60 mg L-1 dose treatment was able to suppress nitrate levels to 0.73±0.03 mg L-1 on the 30th day of cultivation. The addition of nipa husk flour at a dose of 60 mg L-1 was the most effective dose in increasing the abundance of denitrifying bacteria and maintaining the cultivation environment conditions.
