      Imotilisasi Ikan Kaviat (Barbonymus schwanenfeldii) Menggunakan Filtrat Buah Pala dan Metode Kejut Suhu Beku

      Abstract
      Ikan kaviat (Barbonymus schwanenfeldii) merupakan salah satu komoditas ikan hias air tawar ekspor dari Indonesia yang menyumbangkan pendapatan negara. Risiko stres yang mengakibatkan kematian ikan selama transportasi dapat diminimalkan dengan penerapan metode anestesi menggunakan antimetabolit atau suhu rendah. Penelitian bertujuan untuk menentukan konsentrasi anestesi dari filtrat buah pala, penambahan gliserol, waktu kejut suhu beku, dan suhu media pembugaran yang berbeda untuk memperoleh survival rate tertinggi serta pengaruh terhadap kondisi insang ikan kaviat. Metode penelitian meliputi pembuatan filtrat daging buah pala dan pengamatan waktu setiap tahapan pembiusan dari konsentrasi larutan anestesi uji (10%; 12,5%; 15%; 17,5%; dan 20% (v/v)), kemudian perhitungan tingkat kelangsungan hidup dilakukan terhadap ikan dengan pemberian konsentrasi larutan anestesi terpilih, penambahan gliserol 5%, perbedaan waktu kejut beku (10 dan 15 menit) dan suhu media pembugaran (28°C dan 10°C) yang berbeda, dilanjutkan dengan pengamatan kondisi histologis insang dari perlakuan yang menghasilkan tingkat kelangsungan hidup tertinggi. Konsentrasi larutan filtrat daging buah pala terpilih sebagai antimetabolit untuk ikan kaviat adalah 15% (v/v). Penambahan gliserol 5%, waktu kejut suhu beku 10 menit, dan suhu media pembugaran 28°C menghasilkan tingkat kelangsungan hidup yang lebih tinggi, sedangkan kombinasi ketiga faktor tersebut dengan konsentrasi larutan filtrat daging buah pala 15% (v/v) menghasilkan kondisi insang terbaik.
       
      Tinfoil barb (Barbonymus schwanenfeldii) is one of the Indonesian’s exported freshwater ornamental fish. The risk of lethality due to stress experienced by the fishes during the transportation can be reduced by anesthetic method using antimetabolites or low temperature. This study aimed to determine the effect of nutmeg flesh filtrate as natural antimetabolites, glycerol addition, freeze-shock time, and reversal media temperature as fish anesthetic procedure to obtained the highest survival rate and to observed gill condition of tinfoil barb. The research method included nutmeg flesh filtration and solution’s concentrations selection (10%; 12,5%; 15%; 17,5%; and 20% (v/v)) based on the gradual anesthesia stage’s durations, survival rate calculation of the fish anaesthetized by the selected antimetabolite solution’s concentration, 5% glycerol soak, differential freeze-shock time (10 and 15 minutes) and reversal media temperature (28°C and 10°C), included histological condition observation of the gills from the highest-survival rate-resulted treatments. The selected nutmeg flesh filtrate solution’s concentration as antimetabolite for tinfoil barb is 15% (v/v). 5% glycerol addition, 10 minutes-freeze-shock time, and 28°C reversal media temperature resulted in the highest survival rate, while the combination of the three factors with 15% (v/v) nutmeg flesh filtrate solution resulted in the best gill condition.
