      Modifikasi Ganda Hidrolisis Asam dan Annealing terhadap Pati Resisten : Tinjauan Sistematis dan Meta-optimasi

      Abstract
      Pati resisten/resistant starch (RS) memiliki potensi untuk pengembangan pangan fungsional. Teknik modifikasi ganda, yaitu hidrolisis asam/acid hydrolysis (AH) dan annealing (ANN), diterapkan untuk meningkatkan kandungan RS. Namun, penelitian-penelitian sebelumnya memberikan hasil yang bervariasi. Beberapa penelitian melaporkan bahwa teknik modifikasi ganda ini dapat meningkatkan jumlah RS, sementara yang lain melaporkan sebaliknya dengan data yang sebelumnya tidak ditautkan. Oleh karena itu, penelitian ini bertujuan untuk menentukan pengaruh modifikasi ganda AH-ANN terhadap kandungan pati resisten melalui tinjauan sistematis dan pendekatan meta-optimasi. Penelitian dilakukan dalam 3 tahapan yaitu tinjauan sistematik/systematic review (SR), metaanalisis (MA), dan metodologi permukaan respons/response surface methodology (RSM). Analisis data untuk SR didasarkan pada pedoman PRISMA. Luaran utama MA adalah ukuran efek dalam nilai Standardized Mean Difference (SMD), yang dihitung menggunakan metode Hedge’s d untuk model random effect. RSM digunakan untuk mengoptimasi hasil MA menggunakan grafik 3D permukaan respons. Hasil penelitian menggunakan pendekatan-pendekatan ini menunjukkan bahwa modifikasi ganda hidrolisis asam dan annealing memberikan pengaruh terhadap menurunnya kadar pati resisten. Hasil utama SR (n=16 data) menunjukkan bahwa kadar RS menurun (n=10 data atau 62,5%), yang diikuti hasil MA (n=9 data) bahwa AH-ANN menurunkan kadar RS sangat signifikan (p < 0,001; SMD: - 10,608; 95% CI: -13,704 hingga -7,511). Hasil meta-optimasi menunjukkan pada parameter 6 jam hidrolisis asam dan pada 50 ℃ suhu inkubasi annealing, memberikan penurunan terkecil pada kadar RS. Penelitian modifikasi ganda AHANN masih membutuhkan data penelitian eksperimental yang lebih banyak agar dapat menemukan parameter yang optimal untuk meningkatkan kadar pati resisten.
       
      Resistant starch (RS) has potential for the development of functional food. Dual modification techniques, i.e., acid hydrolysis (AH) and annealing (ANN), were applied to raise RS content. However, several studies yielded varying results. Some reported that these techniques could increase the quantity of RS, while others reported otherwise with data that have not previously been linked. Therefore, this current work aimed to determine the effects of dual modification AH-ANN methods on resistant starch content through a systematic review and meta-optimization approach. The study was conducted in 3 stages i.e. a systematic review (SR), metaanalysis (MA), and response surface methodology (RSM). Data analysis for SR was based on PRISMA guidelines. The main output of MA was the effect size in Standardized Mean Difference (SMD) values, calculated using the Hedge’s d method for random-effects models. RSM was used to optimize the result of MA using a 3D surface chart. The results of the study using these approaches showed that the dual modification of acid hydrolysis and annealing had an influence on decreasing resistant starch levels The main result of SR (n=16 data) showed that RS content decreased (n=10 data or 62,5%), which follows the MA result (n= 9 data) that AH-ANN decreased RS very significantly (p < 0,001; SMD: -10,608; 95% CI: -13,704 to -7,511). The meta-optimization results showed that at 6 hours of acid hydrolysis and at 50 ℃ annealing incubation temperature presented the smallest decrease in RS content. Dual modification of AH-ANN still requires broadly more experimental research data in order that it can be adequate to discover the optimum parameters to increase the RS.
