dc.description.abstractSelenoprotein W1 (SEPW1) adalah protein yang mengandung selenium yang memiliki peran penting dalam pertumbuhan dan perkembangan otot melalui perlindungan myoblast dari stres oksidatif. Penelitian ini bertujuan untuk mengidentifikasi keragaman gen SEPW1 dan hubungannya dengan kandungan mineral pada domba. Sampel yang digunakan sebanyak 40 ekor domba yang terdiri dari 10 domba ekor tipis (DET), 10 domba compass agrinak (DCA), 10 domba garut (DG), dan 10 domba jonggol (DJ). Kandungan mineral yang terpilih ditentukan dengan menggunakan metode IK LP-04.10-LT-1.0. Identifikasi polimorfisme gen SEPW1 dilakukan menggunakan Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Ukuran produk amplifikasi adalah 455 bp. Uji chi kuadrat menunjukkan populasi yang diamati berada dalam keseimbangan Hardy Weinberg. Keragaman gen SEPW1 ditemukan pada setiap rumpun domba. Hasil analisis menunjukkan SNP g.53617063 A>G gen SEPW1 berasosiasi secara signifikan (P<0.05) dengan kandungan mineral selenium. Gen SEPW1 berpotensi digunakan sebagai penanda seleksi terhadap kandungan mineral pada dombaid
dc.description.abstractSelenoprotein W1 (SEPW1) is a protein containing selenium that has an important role in muscle growth and development through protecting myoblasts from oxidative stress. This study was aim to identify polymorphism of the SEPW1 gene and its association with mineral content in sheep. The samples used were 40 sheep consisting 10 samples of javanese thin tailed sheep (JTT), 10 samples of compass agrinak sheep (CAS), 10 samples garut sheep (GS), and 10 samples of jonggol sheep (JS). Selected mineral contents were determined using the IK LP 04.10-LT-1.0 method. Identification of SEPW1 gene polymorphism were conducted using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). The size of the amplification product were 455 bp. Chi-square analys showed that the population was in Hardy-Weinberg equilibrium. Polymorphism of SEPW1 gene were found in each breed of sheep. A SNP g.53617063 A>G of the SEPW1 gene were significantly associated (P<0.005) with selenium mineral content. The SEPW1 gene was potential to be used as a selection marker for mineral content in lamb.id
