Soybean oil-calcium soap is soap-protected soybean oil as a source of polyunsaturated fatty acids. Provision of soybean oil-calcium soap can removes negative effects of unsaturated fatty acids on rumen microbes and optimize rumen fermentation. The research was aimed to study the effect of soybean oil calcium soap on ammonia concentration, microbial protein synthesis and the activity of cellulase enzymes in vitro. The experiment was designed in completely randomized block design (RBD) with 3 treatments and 4 block based on time rumen sampling. The treatments were control diet (C), C + 5% soybean oil (M), and C + calcium-soap of 5% soybean oil (S). The results showed that supplementation of calcium-soap soybean oil at level 5% did not significantly affect the pH value, NH3 concentration and microbial protein synthesis. Mean while it could increase the activity of cellulase enzyme.
