kualitas sperma ikan koi selama kriopreservasi Koi fish (Cyprinus rubrofuscus) is an ornamental fish with high economic value
and its production depends on the availability of high-quality sperm. However, the
asynchronous maturation of male and female broodstock gonads can become a
constraint in the breeding process. Sperm cryopreservation offers a potential
solution to this issue. The success of the cryopreservation process is influenced by
the type and dosage of cryoprotectants used. This study aimed to analyze the effect
of adding different doses of honey as an extracellular cryoprotectant to a 10%
dimethyl sulfoxide (DMSO) solution on the quality of koi fish sperm after
cryopreservation. Five treatments were tested: a control, 10% DMSO, and
combinations of 10% DMSO with honey at dosages of 0.1%, 0.5%, and 1%.
Parameters observed included sperm motility, viability, and abnormality after
storage periods of 7, 14, and 30 days. The data were analyzed using ANOVA
followed by Duncan's test. The results showed that the addition of 0.5% honey
produced the highest spermatozoa motility and viability at all storage periods
compared with the other treatments, while the abnormality remained below 20%.
Meanwhile, the use of honey at doses that were too low or too high resulted in lower
spermatozoa quality. Based on these results, honey has the potential to be used as
a natural extracellular cryoprotectant, with a dose of 0.5% being the most effective
treatment for maintaining the quality of koi fish sperm during cryopreservation
