      Abstract
      Ulat sutra eri (Samia cynthia ricini) merupakan penghasil sutra non-murbei yang memiliki nilai ekonomi tinggi, tetapi telurnya bersifat non-diapause sehingga menetas dalam waktu singkat. Kondisi tersebut membatasi fleksibilitas dalam pengelolaan produksi. Penyimpanan dingin dapat digunakan untuk menunda perkembangan embrio, namun berpotensi menurunkan viabilitas telur. Penggunaan metanol sebagai krioprotektan permeabel diharapkan dapat mengurangi kerusakan selama penyimpanan sehingga viabilitas telur tetap terjaga. Penelitian ini bertujuan menganalisis pengaruh konsentrasi metanol dan lama perendaman terhadap viabilitas telur ulat sutra eri setelah penyimpanan dingin selama 7 hari dan 10 hari. Penelitian ini menggunakan rancangan acak lengkap faktorial 3 × 3 dengan tiga konsentrasi metanol (1,5; 2,0; dan 2,5 M) serta tiga lama perendaman (3, 5, dan 7 menit). Telur berumur 48 jam disimpan pada suhu sekitar 10 °C, kemudian diamati durasi penetasan, penundaan penetasan, daya tetas, perkembangan embrio, mortalitas larva, morfometrik larva, serta karakteristik kokon. Hasil penelitian menunjukkan bahwa penyimpanan dingin berhasil menunda penetasan telur, tetapi menurunkan daya tetas dibandingkan dengan kontrol. Konsentrasi metanol berpengaruh nyata terhadap durasi penetasan, penundaan penetasan, dan daya tetas, sedangkan lama perendaman umumnya tidak berpengaruh nyata. Pada penyimpanan selama 10 hari, interaksi antara konsentrasi metanol dan lama perendaman berpengaruh terhadap durasi dan penundaan penetasan. Konsentrasi metanol 2,5 M menghasilkan viabilitas telur yang lebih baik dibandingkan konsentrasi lainnya, sedangkan penyimpanan selama 10 hari memberikan penundaan penetasan yang lebih lama, tetapi diikuti oleh penurunan daya tetas. Perlakuan metanol tidak berpengaruh nyata terhadap mortalitas larva, morfometrik larva, maupun karakteristik kokon. Penelitian ini menunjukkan bahwa efektivitas metanol sebagai krioprotektan dipengaruhi oleh lama penyimpanan telur. Konsentrasi metanol 2,5 M merupakan perlakuan yang paling efektif untuk mempertahankan viabilitas telur selama penyimpanan dingin sekaligus menunda penetasan. Temuan ini dapat menjadi dasar pengembangan teknik penyimpanan telur ulat sutra eri untuk mendukung pengaturan waktu penetasan dan meningkatkan efisiensi budidaya
       
      The eri silkworm (Samia cynthia ricini) is a non-mulberry silk producer with high economic value, but its eggs do not undergo diapause and therefore hatch within a short period of time. This condition limits flexibility in production management. Cold storage can delay embryonic development, but it may reduce egg viability. The use of methanol as a permeable cryoprotectant is expected to reduce damage during storage, thereby maintaining egg viability. This study aimed to analyze the effects of methanol concentration and immersion duration on the viability of eri silkworm eggs after cold storage for 7 and 10 days. The study employed a 3 × 3 completely randomized factorial design with three methanol concentrations (1.5, 2.0, and 2.5 M) and three immersion durations (3, 5, and 7 minutes). Eggs that were 48 hours old were stored at approximately 10 °C; the researchers then observed hatching duration, hatching delay, hatchability, embryonic development, larval mortality, larval morphometry, and cocoon characteristics. The results showed that cold storage successfully delayed egg hatching but reduced hatchability compared to the control. Methanol concentration had a significant effect on hatching duration, hatching delay, and hatchability, whereas soaking duration generally had no significant effect. At 10 days of storage, the interaction between methanol concentration and soaking duration affected hatching duration and hatching delay. A methanol concentration of 2.5 M resulted in better egg viability compared to other concentrations, whereas 10-day storage caused a longer delay in hatching but was followed by a decrease in hatchability. Methanol treatment had no significant effect on larval mortality, larval morphometry, or cocoon characteristics. This study demonstrates that the effectiveness of methanol as a cryoprotectant is influenced by the duration of egg storage. A methanol concentration of 2.5 M was the most effective treatment for maintaining egg viability during cold storage while also inducing hatching delay. These findings can serve as a basis for developing egg storage techniques for the Eri silkworm to support hatching timing control and improve cultivation efficiency
