Penelitian ini bertujuan mengevaluasi aplikasi diet hiperkolesterolemia dengan penambahan propylthiouracil (PTU) pada tikus putih galur Sprague-Dawley. Penelitian menggunakan dua perlakuan, yaitu P0 (pakan basal komersial) dan P1 (diet hiperkolesterolemia + PTU 10 ppm). Parameter yang diamati meliputi profil lipid, hematologi, dan diferensiasi leukosit. Data dianalisis menggunakan uji independent sample t-test. Hasil penelitian menunjukkan P1 mengalami peningkatan kadar kolesterol total, LDL, jumlah leukosit, persentase neutrofil, dan persentase monosit, penurunan persentase limfosit dibandingkan P0 (P<0,05), dan perubahan hispatologi hepar. Sementara itu, kadar trigliserida, HDL, hemoglobin, hematokrit, eritrosit, eosinofil, dan basofil tidak berbeda nyata (P>0,05). Hasil penelitian menunjukkan bahwa pemberian perlakuan berhasil menginduksi kondisi hiperkolesterolemia yang disertai respons inflamasi pada tikus putih galur Sprague-Dawley, sehingga berpotensi digunakan sebagai model hewan preklinis dalam penelitian pengembangan intervensi nutrisi dan pakan pada hewan kesayangan. This study aimed to evaluate the use of a hypercholesterolemic diet supplemented with 10 ppm propylthiouracil (PTU) in Sprague-Dawley rats. Two dietary treatments were applied: P0 (commercial basal diet) and P1 (hypercholesterolemic diet + 10 ppm PTU). Lipid profile, hematological profile, and leukocyte differential count were evaluated, and the data were analyzed using an independent samples t-test. Compared with P0, the P1 group showed significantly higher total cholesterol, LDL, total leukocyte count, neutrophil percentage, and monocyte percentage, significantly lower lymphocyte percentage (P<0.05), and hepatic hispathological changes. No significant differences were observed in triglycerides, HDL, erythrocytes, hemoglobin, hematocrit, eosinophils, or basophils (P>0.05). These findings indicate that P1 successfully induced hypercholesterolemia accompanied by an inflammatory response in Sprague-Dawley rats, suggesting its potential as a preclinical animal model.
