Perluasan budidaya kedelai ke lahan kering iklim kering tropis di Indonesia
menghadapi kendala utama berupa cekaman kekeringan yang menurunkan
produktivitas sekaligus meningkatkan risiko pecah polong. Cekaman pada fase
pembungaan hingga pembentukan polong (R1–R5) merupakan fase kritis yang
sangat menentukan hasil biji dan berpotensi meningkatkan kerentanan terhadap
pecah polong. Berdasarkan hal tersebut, penelitian ini bertujuan untuk
mengevaluasi adaptasi kedelai tropis terhadap cekaman kekeringan dan ketahanan
pecah polong pada fase R1-R5 serta mengevaluasi implikasinya terhadap stabilitas
hasil sebagai dasar pengembangan genotipe adaptif pada lahan kering iklim kering.
Bagian pertama memetakan perkembangan penelitian kedelai tahun 1959
2025 menggunakan analisis bibliometrik berbasis artikel terindeks Scopus. Hasil
menunjukkan bahwa penelitian toleransi kekeringan berkembang pesat sejak tahun
2010, sedangkan penelitian pecah polong masih relatif terbatas. Analisis ko
okurensi memperlihatkan bahwa topik drought stress dan pod dehiscence belum
terhubung secara langsung, sehingga masih terdapat peluang penelitian untuk
mengintegrasikan kedua karakter tersebut.
Bagian kedua mengevaluasi peran gen pod dehiscence1 (pdh1) terhadap
ketahanan pecah polong serta keterkaitannya dengan morfologi polong, komposisi
dinding polong, arsitektur akar, dan parameter genetik. Alel resesif pdh1 berperan
sebagai pengendali utama ketahanan pecah polong dan terbukti berasosiasi sangat
kuat dengan ketahanan pecah polong serta berfungsi sebagai gen mayor sekaligus
perfect marker untuk seleksi. Keberadaan gen long juvenile alel resesif (lj)
memperkuat ketahanan pecah polong. Genotipe tahan pecah polong memiliki
kandungan lignin lebih rendah, kandungan karbohidrat lebih tinggi, serta sistem
perakaran yang lebih berkembang. Karakter pecah polong menunjukkan
heritabilitas arti luas yang tinggi, dan galur AP3, AP18, AP21, AP26, serta AP46
teridentifikasi tahan terhadap pecah polong.
Bagian ketiga mengevaluasi respon genotipe yang membawa pdh1 dan lj
terhadap cekaman kekeringan terkontrol dengan potensial air tanah –1,42 MPa atau
setara 50% kapasitas lapang di screen house. Kelompok genotipe yang membawa
kombinasi pdh1 dan lj seperti AP21 dan AP46 menunjukkan performa hasil lebih
tinggi secara signifikan dibandingkan varietas pembanding. Indeks toleransi
kekeringan seperti geometric mean production (GMP), mean production (MP),
stress tolerance index (STI), dan yield index (YI) secara konsisten mampu
mengidentifikasi genotipe unggul, terlebih lagi terdapat korelasi yang signifikan
dengan hasil biji pada kedua kondisi.
Bagian keempat berfokus pada identifikasi karakter adaptif fisiologis dan
biokimia yang mendasari toleransi terhadap kekeringan. Genotipe toleran memiliki
kemampuan yang lebih baik dalam mempertahankan status air tanaman melalui
mekanisme drought avoidance, yang didukung oleh sistem perakaran yang lebih
berkembang dan regulasi stomata yang lebih efektif. Kemampuan tersebut
tercermin dari nilai leaf relative water content (LRWC), laju asimilasi, bobot kering
tajuk, viabilitas polen, dan kandungan karotenoid yang lebih tinggi dibandingkan
genotipe peka. Akumulasi proline yang lebih rendah pada genotipe toleran
menunjukkan bahwa tanaman tidak memerlukan penyesuaian osmotik yang intensif
karena mampu mempertahankan homeostasis air jaringan. Temuan ini
menunjukkan bahwa peningkatan akumulasi proline tidak selalu mencerminkan
mekanisme toleransi terhadap kekeringan. Rendahnya malondialdehyde (MDA)
dan antosianin mengindikasikan bahwa kemampuan mempertahankan status air
juga menjaga keseimbangan redoks, sehingga pembentukan reactive oxygen
species (ROS) dapat ditekan. Proline dan MDA berkorelasi negatif dan signifikan
dengan hasil biji pada kondisi normal maupun cekaman kekeringan, serta
menunjukkan heritabilitas arti luas yang tinggi, yang mengindikasikan adanya
kontrol genetik yang kuat dan ekspresi fenotipik yang stabil.
Bagian kelima berfokus pada pengujian genotipe di lahan kering iklim kering
tipe E1 terhadap cekaman kekeringan dan pecah polong pada bulan Juli –
November 2024. Penghentian penyiraman selama 29 hari yang diberlakukan dari
10 hari sebelum berbunga mampu menurunkan potensial air tanah hingga -3,89
MPa atau 23,35% KL, menurunkan hasil biji, dan meningkatkan pecah polong.
Genotipe yang membawa pdh1 dan lj seperti AP21, AP18, AP26, dan AP46 tetap
menunjukkan performa hasil lebih tinggi dan penurunan hasil yang lebih kecil, serta
tahan terhadap pecah polong. Cekaman kekeringan juga menurunkan konduktansi
stomata dan transpirasi sebagai upaya konservasi air (avoidance) diikuti dengan
penurunan bobot tajuk dan luas kanopi. Selain itu, cekaman kekeringan
menurunkan kandungan kalium (K) pada polong yang mengindikasikan perubahan
fisiologis yang berkaitan dengan respons hormonal asam absisat yang meningkat.
Bagian keenam mengevaluasi stabilitas hasil biji pada beberapa lingkungan
menggunakan parameter stabilitas non-parametrik. Genotipe yang membawa pdh1
dan lj seperti AP21 dan AP46 merupakan genotipe yang memiliki hasil biji tinggi
dan yang paling stabil berdasarkan parameter Si(6) (stability (6)), TOP, RS (rank of
sum), Si(1), Si(2), TOP, NPi(1) (non-parametric(1)), dan NPi(2). Terdapat enam
parameter yang memiliki korelasi yang signifikan dengan hasil biji, yaitu Si(6),
TOP, NPi(3), dan NPi(4) berkorelasi positif, serta RS dan NPi(2) berkorelasi negatif,
sehingga dapat digunakan sebagai indikator seleksi. Analisis dendogram
menunjukkan bahwa genotipe dengan pdh1 dan lj masuk pada kelompok dengan
hasil tinggi dan stabil, yang menegaskan konsistensi performa lintas lingkungan.
Secara keseluruhan, integrasi alel resesif lj dan pdh1 membentuk mekanisme
kanalisasi genetik yang memungkinkan tanaman mempertahankan produktivitas
pada kondisi cekaman kekeringan dan risiko pecah polong. Alel resesif lj
berkontribusi meningkatkan kapasitas source–sink melalui perpanjangan fase
vegetatif, peningkatan akumulasi biomassa dan karbohidrat, perkembangan sistem
perakaran yang lebih baik, serta kemampuan mempertahankan status air tanaman.
Sementara itu, pdh1 berperan dalam menekan lignifikasi dinding polong sehingga
mengurangi pembentukan tegangan mekanik yang memicu pecah polong, serta
memengaruhi peningkatan auksin yang berkontribusi pada perkembangan akar.
Sinergi kedua gen tersebut menunjukkan bahwa toleransi kekeringan dan ketahanan
pecah polong merupakan bagian dari satu sistem adaptasi yang saling mendukung
untuk mempertahankan produktivitas kedelai pada lahan kering tropis. Soybean cultivation expansion into tropical drylands in Indonesia faces major
constraints due to drought stress, which reduces productivity while increasing the
risk of pod shattering. Drought stress occurring during the flowering to pod
formation stages (R1–R5) represents a critical period that largely determines seed
yield and may increase susceptibility to pod shattering. Based on these
considerations, this study aimed to evaluate the adaptation of tropical soybean to
drought stress and pod shattering resistance during the R1–R5 stages, and to assess
their implications for yield stability as a basis for developing soybean genotypes
adapted to tropical dryland environments.
The first part of this dissertation mapped the development of soybean research
from 1959 to 2025 using a bibliometric analysis based on Scopus-indexed
publications. The results showed that research on drought tolerance has increased
rapidly since 2010, whereas studies on pod shattering remain relatively limited. Co
occurrence network analysis revealed that the topics drought stress and pod
dehiscence are not directly connected, indicating substantial opportunities for
integrating these two research areas.
The second part evaluated the role of the pod dehiscence1 (pdh1) gene in pod
shattering resistance and its association with pod morphology, pod wall
composition, root architecture, and genetic parameters. The recessive pdh1 allele
was identified as the primary determinant of pod shattering resistance, showing a
strong association with the resistant phenotype and functioning as a major gene as
well as a perfect marker for selection. The presence of the recessive long juvenile
(lj) allele further enhanced pod shattering resistance. Resistant genotypes exhibited
lower lignin content, higher carbohydrate content in the pod wall, and more
developed root systems. Pod shattering displayed high broad-sense heritability, and
the advanced lines AP3, AP18, AP21, AP26, and AP46 were identified as resistant
to pod shattering.
The third part evaluated the response of genotypes carrying pdh1 and lj under
controlled drought stress at a soil water potential of -1.42 MPa, equivalent to 50%
field capacity, under screen house conditions. Genotypes carrying the pdh1–lj
combination, particularly AP21 and AP46, produced significantly higher seed yield
than the check varieties. Drought tolerance indices, including geometric mean
production (GMP), mean production (MP), stress tolerance index (STI), and yield
index (YI), consistently identified superior genotypes and showed significant
correlations with seed yield under both normal and drought conditions.
The fourth part focused on identifying the physiological and biochemical
adaptive traits underlying drought tolerance. Drought-tolerant genotypes
maintained plant water status more effectively through a drought avoidance
mechanism supported by a more developed root system and more efficient stomatal
regulation. This response was reflected by higher leaf relative water content
(LRWC), photosynthetic assimilation rate, shoot dry weight, pollen viability, and
carotenoid content than those of drought-sensitive genotypes. Lower proline
accumulation in tolerant genotypes indicated that intensive osmotic adjustment was
unnecessary because tissue water homeostasis was effectively maintained. These
findings suggest that increased proline accumulation does not necessarily represent
a drought tolerance mechanism. Lower malondialdehyde (MDA) and anthocyanin
contents further indicated that maintenance of plant water status also preserved
cellular redox balance, thereby reducing the formation of reactive oxygen species
(ROS). Both proline and MDA were negatively and significantly correlated with
seed yield under normal and drought conditions and exhibited high broad-sense
heritability, indicating strong genetic control and stable phenotypic expression.
The fifth part evaluated soybean genotypes under tropical dryland conditions
(E1 agroclimatic zone) from July to November 2024. Withholding irrigation for 29
days, beginning 10 days before flowering, reduced soil water potential to -3.89
MPa (23.35% field capacity), resulting in lower seed yield and increased pod
shattering. Nevertheless, genotypes carrying pdh1 and lj, including AP18, AP21,
AP26, and AP46, maintained higher seed yield, experienced smaller yield
reductions, and remained resistant to pod shattering. Drought stress also reduced
stomatal conductance and transpiration as part of a water conservation (avoidance)
strategy, accompanied by reductions in shoot dry weight and canopy size. In
addition, drought stress decreased potassium (K) concentration in pods, indicating
physiological changes associated with increased abscisic acid (ABA) signaling.
The sixth part evaluated seed yield stability across multiple environments
using non-parametric stability parameters. Genotypes carrying pdh1 and lj,
particularly AP21 and AP46, consistently exhibited high and stable seed yield based
on Si(6) (stability (6)), TOP, RS (rank of sum), Si(1), Si(2), NPi(1) (non-parametric
(1)), and NPi(2). Six parameters were significantly correlated with seed yield: Si(6),
TOP, NPi(3), and NPi(4) were positively correlated, whereas RS and NPi(2) were
negatively correlated, indicating their usefulness as selection criteria. Cluster
analysis further grouped the pdh1–lj genotypes within the high-yielding and stable
cluster, demonstrating consistent performance across environments.
Overall, the integration of the recessive lj and pdh1 alleles establishes a
genetic canalization mechanism that enables soybean plants to maintain
productivity under drought stress and high pod shattering risk. The recessive lj
allele contributes to enhanced source–sink capacity through prolonged vegetative
growth, higher biomass and carbohydrate accumulation, improved root system
development, and better maintenance of plant water status. Meanwhile, pdh1
reduces pod wall lignification, thereby minimizing the mechanical tension
responsible for pod shattering, while also potentially enhancing auxin accumulation
associated with root development. The synergistic interaction of these two genes
demonstrates that drought tolerance and pod shattering resistance are not
independent traits but rather components of an integrated adaptive system that
supports soybean productivity under tropical dryland conditions.
