      Abstract
      Strategi perancangan percobaan yang selaras dengan pemodelan statistika sangat penting untuk memperoleh informasi yang efisien dan andal dalam studi formulasi. Mixture experiments merupakan rancangan percobaan yang digunakan ketika respons dipengaruhi oleh proporsi relatif dari beberapa komponen penyusun, dengan jumlah seluruh proporsi harus bernilai 1. Pada mixture experiments, metode optimal design banyak digunakan untuk mengurangi jumlah satuan percobaan yang diperlukan sekaligus mempertahankan kualitas informasi statistik yang diperoleh. Penelitian ini berfokus pada percobaan Mixture of Mixtures (MoM), yang memiliki struktur hierarkis dan umumnya memerlukan jumlah satuan percobaan yang besar. Untuk mengatasi keterbatasan strategi optimal design pada percobaan MoM, penelitian ini bertujuan memperoleh rancangan percobaan yang optimal berdasarkan kriteria D-optimality dan I-optimality menggunakan algoritma Variable Neighborhood Search (VNS) dengan strategi multi-start, serta menerapkan rancangan terbaik pada studi kasus formulasi senyawa antioksidan tanaman herbal. Rancangan D-optimal bertujuan memaksimumkan determinan matriks informasi, sedangkan rancangan I-optimal bertujuan meminimumkan ragam prediksi respons dari daerah percobaan. Pada penelitian ini, rank matriks informasi lebih kecil daripada jumlah parameter model, sehingga perhitungan kriteria D-optimality dan I-optimality dilakukan menggunakan pendekatan regularisasi ridge. Penelitian mempertimbangkan dua skenario, yaitu tanpa constraint dan dengan constraint pada komponen, serta berbagai jumlah satuan percobaan yang berkaitan dengan banyaknya parameter model. Kinerja rancangan dievaluasi menggunakan D-efficiency, I-efficiency, dan Fraction of Design Space (FDS) plot untuk membandingkan kualitas estimasi parameter dan kemampuan prediksi pada setiap rancangan yang dihasilkan. Hasil penelitian menunjukkan bahwa pada kondisi tanpa constraint, rancangan D-optimal dan I-optimal menghasilkan struktur rancangan serta kinerja statistik yang serupa. Kedua kriteria menghasilkan distribusi ragam prediksi yang hampir identik ketika jumlah satuan percobaan relatif besar. Namun, ketika jumlah satuan percobaan menurun dibandingkan kompleksitas model, rancangan D-optimal menunjukkan ragam prediksi yang sedikit lebih rendah pada sebagian besar ruang desain. Sebaliknya, pada kondisi dengan constraint, perbedaan antara kedua kriteria mulai terlihat ketika jumlah satuan percobaan cukup besar. Rancangan I-optimal memberikan kinerja prediksi yang lebih baik dengan tetap mempertahankan efisiensi determinan yang tinggi. Akan tetapi, ketika jumlah satuan percobaan relatif kecil, kedua kriteria menghasilkan rancangan dan kinerja prediksi yang hampir identik. Temuan ini menunjukkan bahwa perbedaan antara rancangan D-optimal dan I-optimal tidak hanya dipengaruhi oleh keberadaan constraint, tetapi juga oleh jumlah satuan percobaan yang digunakan. Dengan demikian, pendekatan yang diusulkan memberikan panduan praktis dalam pemilihan kriteria optimalitas pada percobaan MoM, khususnya ketika sumber daya penelitian terbatas. Rancangan yang diperoleh kemudian diterapkan pada formulasi obat herbal. Sebanyak 20 formulasi disiapkan dan diuji secara eksperimental menggunakan metode DPPH (2,2-diphenyl-1-picrylhydrazyl) untuk mengukur aktivitas antioksidan berdasarkan persentase inhibisi (% inhibition), di mana nilai yang lebih tinggi menunjukkan aktivitas antioksidan yang lebih kuat. Hasil pengujian menunjukkan aktivitas antioksidan yang bervariasi, yaitu berkisar antara 28,98% hingga 88,19% inhibisi. Pemodelan menggunakan regresi LASSO memberikan performa yang baik dengan nilai R^2 sebesar 0,8995 dan RMSE sebesar 5,816. Berdasarkan model tersebut, formulasi optimum diprediksi berada pada komposisi mayor (c_1,c_2,c_3 )=(0.1,0.8974,0.0026) dengan nisbah bahan–pelarut (m_1,m_2 )=(0.0909,0.9091), yang menghasilkan prediksi aktivitas antioksidan sebesar 89,55%. Hasil analisis menunjukkan bahwa rancangan optimal yang dihasilkan tidak hanya efisien secara statistik, tetapi juga efektif secara praktis dalam mengidentifikasi formulasi dengan respons tinggi pada percobaan Mixture of Mixtures yang memiliki constraint.
       
      An experimental design strategy aligned with statistical modeling is essential to obtain efficient and reliable information in formulation studies. Mixture experiments are experimental designs in which the response depends on the relative proportions of the mixture components, while the total proportion of all components is constrained to equal 1. In mixture experiments, optimal design methods are widely used nowadays to reduce the number of runs, but remain from a statistical point of view. This study focuses on Mixture of Mixtures (MoM) experiments, which involve hierarchical structures and typically require a large number of experimental runs. To address the lack of optimal design strategies for MoM experiments, this study aims to obtain optimal experimental designs based on the D-optimality and I-optimality criteria using a multi-start Variable Neighborhood Search (VNS) algorithm, and to apply the selected design to a case study of herbal antioxidant formulation. D-optimality seeks to maximize the determinant of the information matrix, whereas I-optimality aims to minimize the average prediction variance. In this study, the rank of the information matrix was smaller than the number of model parameters; therefore, ridge regularization was employed in the computation of the D-optimality and I-optimality criteria. The proposed approach was evaluated under two scenarios, namely unconstrained and constrained component compositions, and across various numbers of experimental runs relative to the number of model parameters. Design performance was assessed using D-efficiency, I-efficiency, and Fraction of Design Space (FDS) plots to compare the quality of parameter estimation and prediction performance of the resulting designs. The results show that, in the unconstrained ingredients, the resulting D- and I-optimal designs produced highly similar structures and statistical performance. The two criteria yielded almost identical prediction-variance distributions for larger run sizes. The D-optimal design exhibited slightly lower prediction variance across much of the design space as the number of runs decreased relative to the model complexity. On the contrary, for constrained ingredients, differences between the two criteria emerged when the number of runs was sufficiently large. The I-optimal design provided improved prediction performance while maintaining high determinant efficiency. However, when the number of runs was small, both criteria yielded nearly identical designs and prediction performance. These findings indicated that the distinction between the resulting D- and I-optimal designs depended not only on the presence of constraints but also on the run size. In this research, the proposed approach provides practical guidance for selecting optimality criteria in MoM experiments, particularly under resource limitations. The design was implemented on a herbal medicine formulation. The formulation of 20 runs were prepared and tested experimentally using the DPPH (2,2-diphenyl-1-picrylhydrazyl) assay to measure antioxidant activity, % inhibition, where higher values indicate stronger antioxidant activity. The experimental results showed a wide range of antioxidant activities, from 28.98% to 88.19% inhibition. LASSO regression provided the best predictive performance, with an R^2 of 0.8995 and an RMSE of 5.816. Based on the fitted model, the optimal formulation was predicted at the major-mixture composition (c_1,c_2,c_3 )=(0.1,0.8974,0.0026) and the minor-mixture composition (m_1,m_2 )=(0.0909,0.9091), yielding a predicted antioxidant activity of 89.55% inhibition. The analysis confirmed that the resulting optimal design is not only statistically efficient but also practically effective for identifying high-response formulations in constrained Mixture of Mixtures experiments.
