      Pengembangan database Near-Infrared Spectroscopy (NIRS) dalam memprediksi VFA parsial pada cairan rumen sapi perah

      Abstract
      Penelitian ini bertujuan untuk mengembangkan database Near Infrared Spectroscopy (NIRS) dalam memprediksi volatile fatty acids (VFA) parsial dari cairan rumen sapi perah sebagai metode alternatif yang lebih cepat dan efisien. Sampel cairan rumen dianalisis menggunakan metode Gas Chromatography (GC) dan NIRS dengan alat Buchi NIRFlex N-500. Spektrum NIRS dikalibrasi dan divalidasi dengan aplikasi NIRWare Management Console dan NIRCal 5.2. evaluasi model dilakukan berdasarkan nilai koefisien determinasi (R²), standard error (SEC dan SEP), residual predictive deviation (RPD), dan Prediction Residual Error Level (PRL). Hasil kalibrasi dan validasi internal menunjukkan nilai R² sebesar 0,67–0,82 dan nilai RPD sebesar 1,52 2,17, hal ini menunjukkan model dapat digunakan untuk screening serta prediksi secara kuantitatif. Validasi eksternal menunjukkan bahwa asam asetat, asam propionat, dan asam butirat tidak berbeda nyata (P>0,05) terhadap hasil GC, sedangkan asam iso butirat, asam iso-valerat, dan asam valerat menunjukkan perbedaan nyata (P<0,05). Simpulan dari penelitian ini yaitu metode NIRS berpotensi digunakan sebagai metode alternatif yang cepat dan efisien dalam pendugaan VFA parsial makro (asam asetat. asam propionat, dan asam butirat) dari cairan rumen sapi perah.
       
      This study aimed to develop a Near-Infrared Spectroscopy (NIRS) database for predicting partial volatile fatty acids (VFA) in dairy cow rumen fluid as a faster and more efficient alternative method. Rumen fluid samples were analyzed using Gas Chromatography (GC) and NIRS with the Buchi NIRFlex N-500 instrument. NIRS spectra were calibrated and validated using NIRWare Management Console and NIRCal 5.2 software. Model evaluation was conducted based on the coefficient of determination (R²), standard error (SEC and SEP), residual predictive deviation (RPD), and Prediction Residual Error Level (PRL). Calibration and internal validation results showed R² values ranging from 0.67 to 0.82 and RPD values ranging from 1.52 to 2.17, indicating that the model could be used for screening and quantitative prediction purposes. External validation showed that acetic acid, propionic acid, and butyric acid were not significantly different (P>0.05) compared to GC results, whereas iso-butyric acid, iso-valeric acid, and valeric acid showed significant differences (P<0.05). In conclusion, the NIRS method has potential as a rapid and efficient alternative method for estimating major partial VFA components (acetic acid, propionic acid, and butyric acid) in dairy cow rumen fluid.
