      Abstract
      Streptococcus pneumoniae merupakan bakteri yang dapat menyebabkan penyakit seperti otitis media, pneumonia, meningitis, bakteremia, dan sepsis serta memperoleh sebagian besar patogenisitasnya dari kapsul polisakarida (CPS). Penelitian ini bertujuan untuk mengkarakterisasi lokus kapsul untuk biosintesis kapsul polisakarida, resistensi antibiotik, dan variasi genetik dari Streptococcus pneumoniae serotipe 15B di Indonesia. Diperoleh bahwa serotipe 15B memiliki prevalensi tinggi di wilayah Lombok, Manado, Sorong, Sumbawa, dan Semarang. Secara genomik, isolat ini memiliki rata-rata konten GC 39,6% dan ukuran genom sekitar 2,1 Mbp, dengan dominasi sequence type ST193 dan ST20231 dalam Global Pneumococcal Sequence Cluster (GPSC) 11. Terdapat sejumlah isolat yang menunjukkan resistensi terhadap kloramfenikol, tetrasiklin, trimetoprim, sulfametoksazol, dan ko-trimoksazol. Variasi genetik dari keseluruhan isolat 15B yang diujikan tergolong rendah, dengan kemiripan mencapai 94%–100%
       
      Streptococcus pneumoniae is a bacterium capable of causing diseases such as otitis media, pneumonia, meningitis, bacteremia, and sepsis also derives most of its pathogenicity from its capsular polysaccharide (CPS). This study aims to characterize the capsule locus, antibiotic resistance, and genetic variation of Streptococcus pneumoniae serotype 15B in Indonesia. The results showed that serotype 15B has a high prevalence in the Lombok, Manado, Sorong, Sumbawa, and Semarang regions. Genomically, these isolates have an average GC content of 39.6% and a genome size of approximately 2.1 Mbp, with a dominance of sequence types ST193 and ST20231 within the Global Pneumococcal Sequence Cluster (GPSC) 11. Regarding drug resistance, some of the isolates showed resistance to chloramphenicol, tetracycline, trimethoprim, sulfamethoxazole, and co-trimoxazole. The genetic variation among all tested 15B isolates was low, with similarity ranging from 94% to 100%
