      Fraksinasi dan Karakterisasi Peptida Bioaktif Putih Telur Ayam IPB-D1 Hasil Hidrolisis Enzim Tripsin sebagai Antioksidan dan Antibakteri

      Abstract
      Putih telur ayam IPB-D1 merupakan produk dari ayam lokal Indonesia dengan komposisi peptida yang unik dan berkualitas tinggi yang memiliki potensi besar sebagai sumber peptida bioaktif. Aktivitas antioksidan dan antibakteri sangat dipengaruhi oleh komposisi asam amino dan muatan ioniknya. Proses fraksinasi seperti kromatografi pertukaran ion diperlukan untuk memisahkan peptida berdasarkan muatannya guna meningkatkan kemurnian dan efektivitas bioaktifnya. Penelitian ini bertujuan melakukan fraksinasi peptida bioaktif putih telur ayam IPB-D1 hasil hidrolisis enzim tripsin melalui kromatografi pertukaran ion serta mengevaluasi aktivitas antioksidan dan antibakterinya. Metode penelitian meliputi pengukuran nilai haugh unit (HU), kadar protein, hidrolisis enzimatis, ultrafiltrasi menggunakan membran <3kDa, fraksinasi menggunakan kromatografi pertukaran ion, aktivitas antioksidan dan aktivitas antibakteri. Rancangan percobaan yang digunakan dalam penelitian ini adalah rancangan acak lengkap (RAL) pola searah. Hasil menunjukkan seluruh fraksi peptida memiliki aktivitas antioksidan dan antibakteri yang lebih baik dibandingkan dengan putih telur tanpa perlakuan, hidrolisat, dan filtratnya. Fraksi 3 menunjukkan aktivitas penangkapan radikal DPPH tertinggi (52,42%) dan kapasitas antioksidan tertinggi (3558,78 mg EVC 100 g?¹), diikuti oleh Fraksi 2 dan Fraksi 4. Selain itu, aktivitas antibakteri terbesar ditunjukkan oleh Fraksi 2 (terhadap bakteri Bacillus cereus (2,52 mm) dan Salmonella typhi (2,82 mm)) dan Fraksi 3 (terhadap bakteri Staphylococcus aureus (1,56 mm) dan Escherichia coli (2,53 mm)). Oleh karena itu, kromatografi pertukaran anion terbukti efektif dalam meningkatkan aktivitas antioksidan dan antibakteri peptida putih telur ayam IPB-D1.
       
      Egg white from IPB-D1 chicken, a local Indonesian breed with a unique and high-quality peptide composition holds great potential as a source of bioactive peptides. Antioxidant and antibacterial activities are highly influenced by amino acid composition and ionic charge. Fractionation processes such as ion exchange chromatography are necessary to separate peptides based on their charge in order to enhance their purity and bioactive effectiveness. This study aimed to fractionate bioactive peptides derived from trypsin-hydrolyzed IPB-D1 egg white using anion exchange chromatography and to evaluate their antioxidant and antibacterial activities. The research methods included measurement of Haugh Unit (HU), protein content, enzymatic hydrolysis, ultrafiltration using a <3 kDa membrane, fractionation using ion exchange chromatography, antioxidant activity, and antibacterial activity. The experimental design used in this study was a completely randomized design (CRD) with a one-factor arrangement. The results showed that all peptide fractions exhibited superior antioxidant and antibacterial activities compared to untreated egg white, hydrolysate, and filtrate. Fraction 3 demonstrated the highest DPPH radical scavenging activity (52,42%) and antioxidant capacity (3558,78 mg EVC 100 g?¹), followed by Fraction 2 and Fraction 4. Furthermore, the largest antibacterial activity were observed in Fraction 2 (against Bacillus cereus (2,52 mm) and Salmonella Typhi (2,82 mm)) and Fraction 3 (against Staphylococcus aureus (1,56 mm) and Escherichia coli (2,53 mm)). These findings indicate that anion exchange chromatography is an effective method to enhance the functional properties of bioactive peptides from IPB-D1 chicken egg white, with potential applications in the development of natural antioxidant and antimicrobial agents.
