      Analisis Keragaman Genetik Benih Hibrida Ikan Lele Sangkuriang dengan Lele Thailand menggunakan Metode PCR-RAPD

      Abstract
      Ikan lele Sangkuriang merupakan hasil perbaikan genetik melalui persilangan ikan lele Dumbo betina generasi 2 (F2) dengan jantan generasi 6 (F6). Hibridisasi ikan lele Sangkuriang dengan ikan lele introduksi asal Thailand dilakukan untuk memperoleh efek heterosis pada benih hibridanya. Penelitian ini bertujuan menganalisis keragaman genetik benih hibrida ikan lele Sangkuriang dengan lele Thailand dari tiga ukuran berbeda menggunakan polymerase chain reaction (PCR) dengan metode RAPD (random amplified polymorphic DNA). Empat primer (OPA13, OPB-08, OPC-19, OPD-02), dari tiga belas kandidat primer yang diuji menghasilkan pita yang dapat di analisis. Persilangan ikan lele Sangkuriang betina dengan ikan lele Thailand jantan terdiri dari empat pasang induk dengan pemijahan semi alami. Sampel benih hibrida dari setiap pasangan induk dipelihara secara terpisah pada kondisi wadah pemeliharaan yang sama. Sampel DNA diambil dari jaringan sirip setiap ulangan hasil persilangan sesuai dengan kategori ukuran besar (B), sedang (S), kecil (K), dan induk (I). Hasil penelitian menunjukkan nilai heterozigositas benih hibrida berkisar 0,167?0,232 dengan tingkat polimorfisme 72,95–93,88%. Nilai jarak genetik terendah ialah 0,001 antara sampel benih B dan K, dan tertinggi ialah 0,018 antara sampel K dan I. Hasil keragaman genetik dari tiga parameter pada penelitian ini dapat menjadi informasi untuk pemuliaan dan produksi benih ikan lele.
       
      Sangkuriang catfish is the result of genetic improvement through the crossing of female Dumbo catfish generation 2 (F2) with male generation 6 (F6). Hybridization of Sangkuriang catfish and introduced catfish from Thailand was conducted to obtain heterosis effect on its hybrid fry. This study aimed to analyze the genetic diversity of hybrid fry of Sangkuriang catfish and Thailand catfish of three different sizes using polymerase chain reaction (PCR) with RAPD (random amplified polymorphic DNA) method. Four primers (OPA-13, OPB-08, OPC-19, OPD-02), out of thirteen candidate primers tested produced bands that could be analyzed. The crossing of female Sangkuriang catfish with male Thailand catfish consisted of four pairs of broodstocks with semi-natural spawning. Hybrid fry samples from each parental pair were reared separately under the same rearing container conditions. DNA samples were taken from the fin tissue of each replicate of the cross according to the size categories of large (B), medium (S), small (K), and broodstock (I). The results showed that the heterozygosity value of hybrid seeds ranged from 0.167-0.232 with a polymorphism level of 72.95-93.88%. The lowest genetic distance value was 0.001 between B and K seed samples, and the highest was 0.018 between K and I samples. The results of genetic diversity of the three parameters in this study can be information for breeding and production of catfish seed.
