| dc.description.abstract | Diare menjadi permasalahan kesehatan utama di dunia. Salah satu penyebab diare adalah infeksi bakteri Enteropathogenic Escherichia coli (EPEC). Penanganan bakteri patogen menggunakan antibiotik sudah tidak lagi efektif karena bakteri dapat meregulasi sistem pertahanannya sehingga mendapatkan resistensi terhadap antibiotik. Bakteriofag sebagai virus yang menyerang bakteri memiliki keunggulan karena memiliki spesifisitas yang tinggi pada tingkat spesies hingga strain suatu bakteri. Penelitian ini bertujuan mengisolasi dan mengkarakterisasi bakteriofag Enteropathogenic E. coli (EPEC) penyebab diare. Bakteriofag diisolasi dan dipurifikasi menggunakan teknik standard double layer. Karakterisasi bakteriofag dilakukan dengan uji kisaran inang, perhitungan nilai Multiplicity of Infection (MOI), dan uji kinetika adsorpsi untuk mengetahui infektivitas dari bakteriofag. Bakteriofag dengan nama FPMY dan FPMA berhasil diisolasi dari air tempat penyimpanan udang di Pasar Ikan Muara Baru, Jakarta Utara. Isolat FPMY dan FPMA menunjukkan morfologi plak yang serupa serta aktivitas litik yang cukup spesifik terhadap kultur EPEC. FPMY memiliki nilai MOI dan kinetika adsorpsi yang lebih tinggi daripada FPMA. | |
| dc.description.sponsorship | Diarrhea is a major global health problem. One of the causes of diarrhea is infection by Enteropathogenic Escherichia coli (EPEC). The treatment of pathogenic bacteria using antibiotics is no longer effective because bacteria regulate their defense systems, leading to antibiotic resistance. Bacteriophages, as viruses that attack bacteria, have the advantage of high specificity at the species level and even at the strain level of bacteria. This study aims to isolation and characterization of bacteriophages of Enteropathogenic E. coli (EPEC) that cause diarrhea. Bacteriophages were isolated and purified using the standard double layer technique. Bacteriophages characterization was subjected with host range tests, calculation of the Multiplicity of Infection (MOI), and adsorption kinetics tests to determine the infection ability of the bacteriophages. Bacteriophages named FPMY and FPMA were successfully isolated from water used for shrimp storage at Pasar Ikan Muara Baru, North Jakarta. The isolates FPMY and FPMA showed similar plaque formation motphology and both had highly-specific lytic activity against EPEC. FPMY had higher MOI values and adsorption kinetics than FPMA. | |
