      Upaya Peningkatan Performa Reproduksi Kima Sisik (Tridacna squamosa) Melalui Perbedaan Dosis Induksi Hormon Serotonin

      Abstract
      Kima sisik (Tridacna squamosa) merupakan salah satu spesies yang dibudidayakan di Indonesia, dengan penjualan sebesar US$ 69,826.-/tahun. Akibat tekanan antropogenik, CITES mengelompokkan kima ke dalam kategori Apendiks II. Penelitian ini bertujuan mengevaluasi kinerja reproduksi T. squamosa yang diinduksi dengan serotonin dosis berbeda terhadap respons pemijahan fekunditas, diameter telur, derajat pembuahan, derajat penetasan, sintasan, abnormalitas, dan kualitas air. Empat perlakuan dengan tiga kali ulangan individu, menggunakan rancangan acak lengkap faktor tunggal yaitu dengan dosis serotonin 0 mM (kontrol) 1,1 mM (P1), 1,4 mM (P2), dan 1,7 mM (P3). Respons pemijahan P1 dan P2 berkisar 45 detik, dan P3 berkisar 60 detik. Persentase memijah tertinggi P2 yaitu 91,67% dan waktu pelepasan sperma tercepat 3 menit 21 detik (P2) dan terlama waktu mendekati 4 menit (P3). Pelepasan telur tercepat berkisar 24 menit (P2) dan terlama berkisar 84 menit (P1), dan menghasilkan telur berdiameter 128,72±1,56 μm (P1, P2). Nilai regresi polinomial fekunditas menghasilkan koefisien determinasi moderat (R² = 0,3885). Induksi serotonin tidak berpengaruh terhadap abnormalitas telur (P>0,1). Derajat pembuahan, derajat penetasan, dan sintasan larva berkisar 70% hingga 80% untuk P1 dan P2. Dosis induksi serotonin yang optimum terhadap kinerja reproduksi T. squamosa yaitu 1,36 mM dan pada dosis lebih tinggi menunjukan pengaruh inhibisi.
       
      Fluted clam (Tridacna squamosa) is one of the clam species cultured in Indonesia, valued around US$ 69.826.-/year Due to anthropogenic pressure, CITES classifies clams into Appendix II. This study evaluates reproductive performances of T. squamosa induced with different doses of serotonin on the parameters, spawning response, fecundity, egg diameter, fertilization rate, hatching rate, survival rate, abnormalities, and water quality. Four treatments with three individual replications, using single factor completely randomized design, with serotonin doses of 0 mM (control) 1.1 mM (P1), 1.4 mM (P2), and 1.7 mM (P3). Spawning response is around 45 seconds (P1, P2), and 60 seconds (P3). Highest spawning percentage was 91.67% (P2), Fastest sperm release was around 3 minutes (P2), and latest was around 4 minutes (P3). Fastest egg release was around 24 minutes (P2) and latest was around 84 minutes (P1), all the eggs diameters were 128.72 ± 1.56 μm (P1, P2). Polynomial regression of fecundity produced moderate coefficient of determination (R² = 0.3885). Egg abnormalities weren’t affected (P>0.1). FR, HR, and SR values ranged from 70% to 80% for P1 and P2. The optimum dose of serotonin induction is 1.36 mM and at higher dosage shows negative feedback.
