      Produksi Ikan Mas Koki (Carassius auratus) Tetraploid dengan Pemberian Kejut Suhu Dingin (Cold Shock)

      Abstract
      Rekayasa set kromosom (poliploidisasi) merupakan salah satu metode untuk memperbaiki produktivitas suatu organisme target dalam mengatasi permasalahan pertumbuhan ikan yang relatif lambat. Tetraploidisasi merupakan metode manipulasi set kromosom pada ikan yang memiliki dua set kromosom (diploid) untuk menghasilkan ikan yang memiliki empat set kromosom (tetraploid). Penelitian ini bertujuan untuk mengevaluasi tetraploidisasi menggunakan kejutan suhu dingin yang berbeda terhadap ikan mas koki. Induksi tetraploidisasi dengan kejutan suhu dingin pada telur ikan mas koki yang telah terbuahi dilakukan pada suhu 8 ℃ (P1), 12 ℃ (P2), dan 16 ℃ (P3) diberikan pada menit ke-29 setelah pembuahan selama 10 menit. Derajat pembuahan telur ikan mas koki berkisar 71,81–75,18%. Tetraploidisi pada ikan mas koki dengan pemberian kejutan suhu dingin menghasilkan derajat penetasan telur berkisar 3,16–5,70% dan abnormalitas larva 13,32–23,24%. Pemberian kejut suhu 8 ℃ selama 10 menit pada zigot berumur 29 menit menghasilkan persentase ikan tetraploid sebesar 30% dan tingkat kelangsungan hidup 76,76%.
       
      Polyploidization is one method to improve the productivity of a target organism in overcoming the problem of relatively slow fish growth. Tetraploidization is a method of manipulating chromosome sets in fish that have two sets of chromosomes (diploid) to produce fish that have four sets of chromosomes (tetraploid). This study aimed to evaluate tetraploidization using different cold temperature shocks for goldfish. Induction of tetraploidization by cold shock on goldfish eggs that had been fertilized at 8 ℃ (P1), 12 ℃ (P2), and 16 ℃ (P3) were administered at 29 minutes after fertilization for 10 minutes. The degree of fertilization of goldfish eggs ranged 71,81–75,18%. Tetraploidy of goldfish with cold shock resulted in hatching rates ranging from 3,16–5,70% and larval abnormalities 13,32–23,24%. Giving a temperature shock of 8 ℃ for 10 minutes to zygotes aged 29 minutes resulted in a percentage of tetraploid fish of 30% with survival rate of 76,76%.
