dc.description.abstractHipertensi penyakit tidak menular tapi banyak menimbulkan resiko timbulnya penyakit lain. Prevelensi hipertensi terus meningkat dari tahun ke tahun begitupun di indonesia, pengobatannya selama ini menggunakan obat sintetik dan menimbulkan efek samping yang merugikan. Adanya dampak yang merugikan tersebut kemudian direspon oleh para peneliti dengan observasi terkait senyawa alami yang dapat menurunkan tekanan darah tinggi melalui jalur penghambatan enzim ACE, salah satunya seperti peptida bioaktif. Peptida bioaktif diperoleh dari berbagai induk protein, tapi proses yang dilakukan berbeda setiap peneliti berbeda dan juga proses produksi peptida bioaktif dipengaruhi banyak faktor, sehingga perlu penjelasan secara menyeluruh hingga dilakukan penarikan kesimpulan terkait hal ini. Berdasarkan latar belakang tersebut maka dilakukan studi sistematik reviu dan meta-analisis agar dapat memberikan gambaran, rekomendasi dan referensi terkait kondisi optimum hidrolisis gelatin ikan laut dalam memproduksi peptida penghambat ACE. Beberapa penelitian terkait hidrolisis enzimatis dalam menghasilkan aktivitas penghambat ACE dari gelatin ikan telah dikumpulkan pada penelitan ini. Metode yang digunakan adalah sistematik reviu dan meta-analisis. Hasil dari studi ini didapatkan optimasi proses produksi peptida bioaktif gelatin ikan. Pada tahap pra- perlakukan direkomendasikan menggunakan asam kuat (24 jam, suhu ruang) atau basa kuat (24-72 jam, suhu 5oC-suhu ruang), tahap ekstraksi perendaman dengan aquades selama 30 jam dengan suhu 60oC dan tahap pemurnian menggunakan sentrifugasi. Hidrolisis enzimatis menggunakan enzim dari mikroba seperti alkalase direkomendasikan. Sistem hidrolisis yang direkomendasikan adalah kombinasi enzim. Karakteristik peptida gelatin ikan penghambat ACE yang didapatkan memiliki berat molekul berkisar 186-829 Da dan susunan rantainya didominasi asam amino hidrofobik. Hasil meta-analisis menunjukan hidrolisis kombinasi enzim lebih baik dari hidrolisis tunggal dengan nilai ukuran efek sebesar 0,593 dan sistem hidrolisis kombinasi yang menghasilkan aktivitas penghambatan ACE tinggi adalah one pot two enzyme.id
dc.description.abstractHypertension is a non-communicable disease but causes a lot of risk for other diseases. The prevalence of hypertension continues to increase from year to year as well as in Indonesia, the treatment has been using synthetic drugs and causing adverse side effects. The existence of these adverse effects was then responded to by researchers with observations related to natural compounds that can reduce high blood pressure through the inhibitory pathway of the ACE enzyme, one of which is bioactive peptides. Bioactive peptides are obtained from various parent proteins, but the process carried out is different for each researcher and the production process for bioactive peptides is influenced by many factors, so a thorough explanation is needed to conclude this matter. Based on this, a systematic review and meta-analysis were carried out to present an overview, recommendation, and reference regarding the optimum conditions for hydrolysis of marine fish gelatin in producing ACE inhibitory peptides. Several studies related to enzymatic hydrolysis in producing ACE inhibitory activity of fish gelatin have been collected in this study. The method used is a systematic review and meta-analysis. The results of this study obtained optimization of the production process of fish gelatin bioactive peptides. In the pre- treatment stage, it is recommended to use strong acid (24 hours, room temperature) or strong base (24-72 hours, 5oC-room temperature), the extraction stage is soaked with distilled water for 30 hours at a temperature of 60oC and the purification stage is using centrifugation. Enzymatic hydrolysis using enzymes from microbes such as alkalase better, for the recommended hydrolysis system is a combination of enzymes. The characteristics of ACE inhibitory fish gelatin peptides obtained have molecular weights ranging from 186-829 Da and the chain arrangement is dominated by hydrophobic amino acids. The results of the meta-analysis showed that the combination of enzyme hydrolysis was better than single hydrolysis with an effect size value of 0.593 and the recommended combination hydrolysis system was one pot two enzymes.id
