      Performa, Kualitas Daging, Profil Usus dan Profil Darah Ayam Broiler yang Diberikan Asam Guanidino Asetat: Meta-Analisis

      Abstract
      Asam guanidino asetat atau guanidinoacetic acid (GAA) dibentuk oleh asam amino arginin dan glisin di organ ginjal yang dikatalisis oleh enzim L-arginine: glycine amidinotransferase. GAA dimetilasi oleh enzim s-adenosyl methionine dan dikonversi menjadi kreatin pada organ hati, kemudian dideposit di otot untuk suplai energi. Kreatin akan diubah menjadi kreatinin yang bersifat irreversible. Kreatin hanya dapat ditemukan pada bahan baku asal hewan namun dalam jumlah yang sangat sedikit dan fluktuatif. Biosintesis kreatin dari GAA juga bisa terganggu jika ayam terkena stres panas. Oleh karena itu, kebutuhan kreatin pada tubuh harus terpenuhi secara terus menerus. Penelitian meta-analisis ini bertujuan mengevaluasi pengaruh level GAA dalam meningkatkan performa, perolehan karkas, kualitas daging, profil usus dan profil darah pada ayam broiler. Studi meta analisis ini dilakukan dengan mengintegrasikan 28 artikel dari berbagai jurnal. Dosis suplementasi GAA dari 0 sampai dengan 8000 ppm. Metode model campuran digunakan dengan level GAA dan strain broiler sebagai efek tetap, sedangkan perbedaan studi digolongkan sebagai efek acak. Jumlah ulangan pada setiap studi dijadikan faktor pembobotan dalam pembuatan model. Berdasarkan hasil penelitian menunjukkan bahwa level GAA dapat meningkatkan rataan pertambahan bobot badan harian (ADG) dan menurunkan konversi pakan hari 0˗21, 0˗35 dan 0˗42 dengan pola kuadratik. Peningkatan level GAA diikuti dengan peningkatan potongan karkas bagian dada, persentase susut masak, tinggi vili pada usus bagian duodenum, kreatin, limfosit dan serum nitrogen oksida. Kenaikan level GAA dapat menurunkan variabel crypt depth pada bagian jejenum, hematokrit, heterofil, dan rasio heterofil terhadap limfosit (H/L). Secara umum dapat disimpulkan bahwa suplementasi GAA pada pakan dapat meningkatkan performa ayam broiler. Hasil interaksi antara level GAA dan strain broiler menunjukkan bahwa strain persilangan Hubbard x Cobb 500 lebih responsif terhadap kenaikan ADG hari 0˗21 dibandingkan Ross 308 dan Arbor Acres. Kenaikan level GAA dapat menurunkan susut masak pada strain Arbor Acres dan Partridge-Shank, tetapi meningkatkan susut masak pada strain Ross 708 dan Ross 308. Strain Ross 308 lebih responsif terhadap peningkatan deposisi kreatin dan penurunan hematokrit. Selain itu, strain Ross 708 lebih responsif terhadap peningkatan serum NO dan penurunan rasio H/L. Saran dari penelitian ini adalah diperlukan studi lebih lanjut mengenai perbedaan jenis kelamin sebagai salah satu konstrain. Hal ini dikarenakan ayam jantan dan betina memiliki karakteristik pertumbuhan yang berbeda. Selain itu perlu dilakukan analisis juga terhadap efek penambahan asam guanidino asetat pada pakan yang mengandung dan tidak mengandung bahan baku asal hewan.
       
      Guanidinoacetic acid (GAA) is formed by arginine and glycine that catalyzed by L-arginine:glycine amidinotransferase in the kidney. In the liver, GAA is methylated by s-adenosyl methionine and converted to creatine, then it deposited into muscle as energy supply. Creatinine will be converted into creatinine, irreversibly. Creatine can only be found in animal-derived raw materials, but in small and fluctuating amounts. The biosynthesis of creatine from GAA can also be impaired if broilers are exposed to heat stress. Therefore, the need for creatine in the body must be continuously met. This study was aimed to evaluating the effect of GAA levels on improving performance, carcass yield, meat quality, gut profile and blood profile in broiler. This meta-analysis was done by integrating 28 articles from various journals. Supplementation doses ranged from 0 to 8000 ppm. The mixed model methodology was employed with GAA level and broiler strain as fixed effects and different studies as random effects. The number of replications in each study was used as a weighting factor in making the model. The results showed that increasing GAA level improved average daily gain and reduced feed conversion ratio day 0˗21, 0˗35 and 0˗42 in a quadratic pattern. A higher GAA also accompanied by increasing variable breast yield, cooking loss, villus height in duodenum, creatine, lymphocyte and nitric oxide (NO) serum. A higher GAA reduced variable crypt depth in jejenum, hematocrit, heterophil and heterophil to lymphocyte (H/L) ratio. It was concluded that supplementation of GAA in feed improved performance of broiler. The results of the interaction between GAA level and broiler strain showed that the Hubbard x Cobb 500 cross strain was more responsive to the increasing ADG days 0˗21 than Ross 308 and Arbor Acres. Increasing GAA level decreased cooking loss in Arbor Acres and Partridge-Shank, but increased cooking loss in Ross 708 and Ross 308. Ross 308 was more responsive to increasing creatine deposition and decreasing hematocrit. In addition, Ross 708 was more responsive to increasing NO serum and decreasing H/L ratio. Suggestions from this research is that further studies regarding sex differences as a constraint are needed. This is because male and female broilers have different growth characteristics. In addition, it is necessary to analyze the effect of adding guanidino acetic acid to feed with and without animal-derived raw materials.
