dc.description.abstractOlive (Olea europaea L) is a native plant of Mediteranian region. This plant is largely cultivated and a major commodity that plays an important role in the economy of that region. Olive plant propagation is still using conventional technique by stem cutting and seeds. The stem cutting method has low efficiency rate and requires high control of enviromental conditions. In vitro propagation can be used as an alternative to produce olive plants rapidly and more efficiently. The aims of this research are to know the most effective medium and BAP plant growth regulator concentration for the growth of olive nodal segment culture and to find a better method of explant sterilization. Sterilization methods consisted of four types of treatment that were a standard surface sterilization, the application of HgCh, chloramphenicol, or Plant Preservative Mixture (PPM) in the culture medium. The most effective sterilization method was the standard sterilization plus PPM. The basic media used were DKW or WPM, and the BAP concentrations were 0, 1, 2, and 4 ppm. The result of this research showed that DKW medium without BAP (0 ppm) was the most effective formulation for the growth of olive nodal segment.id
