dc.description.abstractRice variety IR64 is categorized as a sensitive variety to Aluminum (Al) toxicity. Development of Al-tolerant rice varieties can be done by inserting Al-tolerant gene, which one of them is B11 gene that has been succesfully isolated from Al-tolerant rice genotipe, Hawara Bunar. The B11 gene has been succesfully introduced to plant rice variety IR64 through in-planta technique and produced T1 generation. The objective of this research was to evaluate the stability of B11 gene insertion on T1, T2, and T3 generation derived from IR64 transformed variety through antibiotic selection, molecular analysis, and plant phenotipic observation. Antibiotic selection was done using hygromycin antibiotic with concentration of 20 mg/l. Gene insertion analysis was done with Polymerase Chain Reaction (PCR) technique. The result showed that B11 gene had been integrated in T0, T1, and T2 generation of IR64 transgenic plant. The B11 gene integration in T0 did not cause a chimera. The antibiotic selection showed that antibiotic-tolerant and sensitive ratio of T1 and some T2 and T3 populations was 3:1. Some of the T2 and T3 generation populations showed homozigous on B11 gene.
