Outer membrane protein (OMP) gene is a potential gene to identify Candidatus liberibacter, patogent for CVPD (Citrus Vein Phloem Degeneration) disease in citrus plant. The objectives of this research are to clone and to characterize OMP gene fragment of Indonesian isolate C.liberibacter. Characterization based on nucleotide and amino acid. Cloning of DNA fragment of C.liberibacter OMP gene which isolated from infectious citrus, was ligated to pGEM-T Easy vector and was transformed to E.coli DH5α. DNA isolation showed fragment is 696 bp. Cloning gene fragment showed by growth two single colonies and 696 pb gene fragment from colony PCR and plasmid PCR. Characterization based on nucleotide showed OMP gene 99% homolog with C.liberibacter asiaticus. While amino acid characterization showed relative with outer membrane protein in C.liberibacter africanus, C.liberibacter americanus, C.liberibacter solanacearum, C.liberibacter crescens, and Rhizobium etli. The result of amino acid characterization showed there are conserved protein domain.
