Agarwood oil is often used in medicine, perfume, and cosmetic industries. This study aims to determine the optimum soaking time of agarwood kemedangan of inoculated agarwood trees to isolate the essential oils and to analyse the chemical components by using gas chromatograph-mass spectrometer (GC-MS). The agarwood optimum soaking time was 21 days, giving 0.22−0.92% of yield. Compounds identification by using GC-MS indicated the presence of sesquiterpenes in all agarwood oil. Amounts of sesquiterpenes in natural and inoculated trees did not vary, containing 9−15 compounds. Agarwood oil contain 3−6 marker compounds. Inoculated trees producing agarwood oil have similar quality, but less yield than natural agarwood.en
