This experiment was aimed to evaluate sub-culture level of tissue culture propagation of kepok Unti Sayang banana that meet quality standard of seedling effective cost. This experiment was performed with the Complete Random Design with 6 levels and unequal repetitions, in which the mother plant was the control. The data resulted from this experiment was divided into two types: differences among sub-cultures and the ones between two populations of male flowers. The F-test was used to make intra sub-culture comparison, and proceeded by the DMRT at 5 % when the result was significantly different. The T-student test was used to compare two different populations of the male flowers. The differences among sub-cultures indicate that somaclonal variations had been occured by the existance of male flowers in the 4th and 6th sub-culture. The productions of kepok Unti Sayang were also declined by the increasing frequency of sub-cultures. The differences between two populations of male flowers showed that the population had male flowers got a higher variation than the population which didn’t have male flowers. The population that didn’t have male flowers got no rachis and bracts.en
