Melamine is an organic compound in the form of white crystals and a bit difficult to dissolve in water. Method validation is a procedure that is used to prove the results of an analytical method is valid, accurate and precise. Parameter validation methods include system suitability, linearity recovery, limit of detection (LOD) and limit of quantification (LOQ). Liquid Chromatography-Mass Spectrofotometry (LCMS) analysis method with a standard melamine has good accuracy and precision with a relative standard deviation value of 1.18% (system suitability test) and the mean recovery generated by 89.13 ± 9.47%. Limit of detection (LOD) and limit of quantification (LOQ) obtained respectively at 5 ppb and 7 ppb, linearity with line equation y = 71,343,428 + 1371748x and correlation coefficient (r) 0.9367. The result of BSLT get LC50 are in range 0.02-0.1 ppm (0.02 ppm ≤ LC50 ≤ 0.1 ppm) and if using microsoft excel program LC50 obtained at 0.036 ppm or 0.036 μg / mL. LC50 values showed that was toxicen
