dc.description.abstractQuality of temu lawak rhizome as a medicinal plant, was determined by the content of metabolites, which are influenced by geography and environment where the plant grow. Observation on metabolite diversity of ethanol extract of temu lawak rhizome from five differrent regions (Sragen, Bogor, Karanganyar, Sukabumi, and Wonogiri) in this research was conducted by metabolomic approaches using liquid chromatography-mass spectrometry (LC-MS). Multivariate analysis PCA produced 3 groups of samples based on the region of origin: group 1 (Sragen and Bogor), group 2 (Sukabumi and Wonogiri), and group 3 (Karanganyar). Bioactivity of the rhizome was evaluated by brine shrimp lethality test method to determine their potential as anticancer. Rhizome group 1 had the lowest potential anticancer and could be distinguished from the other samples due to their influence variables value of m/z 249.964, which could not be identified due to the limited database. Group 2 and 3 contained metabolite biomarker at m/z 368.115, which is predicted as curcumin and cyclocurcumin. Sample group 3 had the best potential as anticancer, with an LC50 at 25.04 μg/mL.en
dc.description.abstractKualitas rimpang temu lawak sebagai tanaman obat ditentukan oleh kandungan metabolit, yang dipengaruhi oleh kondisi geografis dan lingkungan tumbuh tanaman. Pengamatan keragaman metabolit ekstrak etanol rimpang temu lawak yang berasal dari 5 daerah berbeda (Sragen, Bogor, Karanganyar, Sukabumi, dan Wonogiri) pada penelitian ini dilakukan dengan pendekatan metabolomik menggunakan kromatografi cair-spektroskopi massa (LC-MS). Analisis komponen utama menghasilkan 3 kelompok sampel berdasarkan daerah asalnya: kelompok 1 (Sragen dan Bogor), kelompok 2 (Sukabumi dan Wonogiri), dan kelompok 3 (Karanganyar). Bioaktivitas rimpang diuji dengan metode uji letalitas larva udang untuk mengevaluasi potensinya sebagai antikanker. Rimpang kelompok 1 memiliki potensi antikanker terendah dan terbedakan dari kelompok sampel lainnya akibat pengaruh variabel nilai m/z 249.964, yang belum dapat diidentifikasi karena keterbatasan database. Rimpang kelompok 2 dan 3 mengandung senyawa penciri dengan sinyal m/z 368.115, yang diduga sebagai kurkumin dan siklokurkumin. Sampel rimpang kelompok 3 memiliki potensi antikanker terbaik dengan LC50 25.04 μg/mL.
