Rancang Bangun Penghitung Koloni Selektif Berdasarkan Pigmen Fluoresein pada Pseudomonas aeruginosa
Abstract
The Pseudomonas aeruginosa is phatogen bacteria, easyly grown in simple media, and included in Standar Nasional Indonesia (SNI) 2006 for packed drinking water. These bacteria can be identified by using genetical test, biochemical test, and pigment induction. Genetical and biochemical test need complicated procedure, differ from pigment induction. This pigment is a biological signal for bacteria existency. Pseudomonas aeruginosa can produce both fluorescein pigment which make it different from other bacteria and pyocyanin which can be distinguished from other species in Pseudomonas genus. The purpose of this research is to clarify this fast method on local isolate of P. aeruginosa and design of colony counter for selective quantification for this fluorescence bacteria. Local bacteria were isolated from packed drinking water using milipore 0.45 μm, then it was tested using biochemical kit, and pigment induction. Quantification of colony is supported by colony counter tool, consist of hardware and program. Program was made from Visual Basic with method of plotting used 24 bit used pointer and method of quantification used cell simulation. Biochemical test trust level of P. aeruginosa was 97 % and nitrase was absent. Fluorescence absorbance wave length was 370 nm and its emission is 508 nm. The precision of colony quantification was 88.93% using sampling and/or pointing colour.
