dc.description.abstractSalam (Syzygium polyanthum Wight.) leaves contain flavonoid compounds that have antioxidant activity. Flavonoid extract of salam leaves was expected to induce the formation of petite colony of yeast (Saccharomyces cereviceae) cells. Induction of petite colonies was one of the way to induce apoptosis in the development of anticancer phytopharmaca. This study aimed to test the effectiveness of flavonoid extract of salam leaves in the induction of apoptosis through the formation of petite colonies of yeast cells and DNA fragmentation test. Yeast cells were incubated by flavonoid extract of salam and visualized by spread plate method. DNA fragmentation test was determined through electrophoresis gel agarose method. Crude extract of flavonoid (F) and fractionation flavonoid extract (F1) at concentration of 10 ppm resulted petite frequency of 100% and 98.96%. Pure flavonoid extract (F1.5) at concentration of 0.1 ppm had resulted a petite frequency of 99.125%. Electroforegram profile in F and F1 also showed a thin smear as an indication of DNA degradation. Numbers of petite colonies induced by the concentration of F1.5 at 0.1 ppm (62 colonies) were ~43% less than the amount induced by F (140 colonies) and F1 (146 colonies). This observation indicated an increase cytotoxicity with increasing purity of extract.en
dc.description.abstractDaun salam (Syzygium polyanthum Wight.) mengandung senyawa flavonoid yang memiliki aktivitas antioksidan. Ekstrak flavonoid daun salam diharapkan mampu menginduksi pembentukan koloni petite sel khamir (Saccharomyces cereviceae). Induksi koloni petite merupakan salah satu cara menginduksi apoptosis dalam pengembangan fitofarmaka antikanker. Penelitian bertujuan untuk menguji efektivitas ekstrak flavonoid daun salam dalam menginduksi apoptosis melalui pembentukan koloni petite yang didukung dengan uji fragmentasi DNA. Sel khamir diinkubasi dengan ekstrak flavonoid daun salam, lalu divisualisasi dengan metode cawan sebar. Uji fragmentasi DNA dilakukan dengan metode isolasi DNA dan elektroforesis gel agarosa. Ekstrak kasar flavonoid (F) dan ekstrak flavonoid terfraksinasi (F1) konsentrasi 10 ppm menghasilkan frekuensi petit sebesar 100% dan 98.96%. Fraksi murni flavonoid (F1.5) konsentrasi 0.1 ppm menghasilkan frekuensi petit sebesar 99.125%. Profil elektroforegram pada F dan F1 juga menunjukkan adanya smear tipis sebagai indikasi adanya degradasi DNA. Jumlah koloni petit yang terinduksi oleh F1.5 pada konsentrasi 0.1 ppm (62 koloni) ~43% lebih sedikit daripada jumlah yang diinduksi oleh F(140 koloni) dan F1(146 koloni). Hal tersebut mengindikasikan adanya peningkatan sitotoksisitas dengan bertambahnya kemurnian ekstrak.
