      Abstract
      Stevia is a plant source of natural sweetener containing stevioside which is 300-400 times sweeter than sucrose. The need for stevia planting materials in a large numbers requires fast methods of plant propagation, one of which is the technique of in vitro culture. The objective of this study was to determine the effect of different vessel culture closures and water immersion on the growth of Stevia rebaudiana Bertoni M. plantlets in vitro. The plant material used was small cuttings of stevia from steril plantlets. The medium was Murashige and Skoog, added with 5 μM BA, 2 μM IAA, and 0,35 μM paclobutrazol. The vessel closures used were screw cap, plastic wrap, and aluminum foil. The culture bottles were placed on plastic tray containing 1 cm-high of water as immersion treatment. The plant growth parameters measured were plantlet height, stem diameter, shoot number, node length, leaf color, leaf area, fresh weight and dry weight after 4 weeks of culture. The data were analyzed with SPSS and followed by Duncan test at a significance level of 5%. Microenvironmental parameters were temperature and light intensity. The results showed that culture vessel cover and water immersion affected significantly the growth of Stevia rebaudiana plantlets. The best growth of plantlets was found on the treatment of aluminum foil closure without water immersion, indicated by the highest plantlets, longest stem node, greener leaf color and biggest leaf area. The treatments of screw caps and plastic wrap having higher light intensity than aluminum foil, gave shorter plantlets, larger stem diameter, and higher shoot numbers. Immersion of the vessels decreased the temperature inside the vessel, and increased the shoot numbers.
       
      Stevia merupakan tanaman sumber pemanis alami yang mengandung senyawa steviosida dengan kadar kemanisan 300-400 kali sukrosa. Kebutuhan akan bibit stevia dalam jumlah besar memerlukan metode perbanyakan tanaman dalam waktu yang singkat, salah satu cara ialah teknik kultur in vitro. Penelitian ini bertujuan mengetahui pengaruh jenis penutup botol kultur dan perendaman air terhadap pertumbuhan planlet Stevia rebaudiana Bertoni M. Bahan tanaman yang digunakan adalah stek kecil in vitro stevia. Media yang digunakan adalah Murashige and Skoog, ditambah BA 5 μM, IAA 2 μM, dan paclobutrazol 0,35 μM. Pucuk stevia ditanam pada botol kultur dengan perlakuan tutup botol ulir, plastic wrap, dan aluminum foil. Perlakuan perendaman dilakukan dengan meletakkan botol kultur pada wadah baki plastik yang berisi air setinggi 1 cm. Parameter pertumbuhan planlet yang diukur ialah tinggi planlet, diameter batang, jumlah tunas, panjang ruas, warna daun, luas daun, bobot basah, dan bobot kering setelah masa kultur 4 minggu. Data dianalisis dengan SPSS dan dilanjutkan dengan uji Duncan pada taraf nyata 5%. Parameter lingkungan mikro ialah suhu dan intensitas cahaya. Hasil penelitian menunjukkan bahwa jenis penutup botol kultur dan perlakuan perendaman air mempengaruhi secara nyata pertumbuhan planlet Stevia rebaudiana. Pertumbuhan planlet terbaik dihasilkan dari perlakuan penutup botol aluminum foil tanpa perendaman air, yaitu untuk tinggi planlet, panjang ruas, warna, dan luas daun. Perlakuan tutup ulir dan plastic wrap dengan intensitas cahaya yang lebih tinggi dari aluminum foil, cenderung menghasilkan planlet lebih pendek, kekar (diameter lebih besar), dan jumlah tunas lebih tinggi. Perendaman mengakibatkan penurunan suhu, yang berpengaruh pada peningkatan jumlah tunas.
