Two of many potential sources of natural materials as an α-glucosidase inhibitors are sambiloto and brotowali. The aims of the study were to obtain the best formula of sambiloto and brotowali extract with highest α-glucosidase enzyme inhibition including its fingerprint information. Extractions were carried out using maceration and ultrasonication methods with water, ethanol 30%, and 70% as the solvents. Sambiloto ultrasonication extract and brotowali maceration extract using ethanol 70% as solvent showed the lowest IC50 value, respectively 55.36 and 68.28 ppm. Both of extracts were then formulated. Formula of sambiloto and brotowali with the concentration ratio of 2 IC50:1 IC50 was the best formula with inhibition value 28.11%. In thin layer chromatography separation, ethyl acetate was the optimum mobile phase for sambiloto extract and chloroform: ethanol: ethyl acetate (0.487: 0: 0.513) is the optimum mobile phase composition for brotowali extract. Ethyl acetate used as mobile phase in formula separation. Formula analysis using high performance liquid chromatography technique with optimum separation condition of sambiloto extract did not show components from brotowali extract, so the separation condition of the formula still need to be optimized. di antaranya adalah sambiloto dan brotowali. Penelitian ini bertujuan memperoleh formula ekstrak sambiloto dan brotowali dengan penghambatan kerja enzim α-glukosidase tertinggi dan informasi sidik jari formula tersebut. Metode ekstraksi yang digunakan adalah maserasi dan ultrasonikasi dengan pelarut air, etanol 30%, dan 70%. Ekstrak sambiloto ultrasonikasi dan ekstrak brotowali maserasi menggunakan etanol 70% menunjukkan nilai IC50 terendah, berturut turut 55.36 dan 68.28 ppm. Formula ekstrak sambiloto dan brotowali dengan nisbah konsentrasi 2 IC50:1 IC50 merupakan formula paling baik dengan inhibisi 28.11%. Pada analisis kromatografi lapis tipis, fase gerak optimum pemisahan komponen sambiloto yaitu etil asetat sedangkan untuk ekstrak brotowali adalah campuran kloroform:etanol:etil asetat (0.487:0:0.513). Pemisahan komponen formula dilakukan menggunakan fase gerak tunggal etil asetat. Analisis formula menggunakan kromatografi cair kinerja tinggi dengan kondisi pemisahan ekstrak sambiloto tidak menunjukkan komponen yang berasal dari ekstrak brotowali, sehingga masih perlu dilakukan pengoptimuman kondisi pemisahan formula.
