Antidiabetic activity and analysis of phytochemistry aqueous and ethanol extract of purple leaf (Graptophyllum pictum (L.) Griff)

dc.description.abstractplants that are in one family has reported to have antidiabetic activity through the mechanism of inhibition of the α-glucosidase enzyme, thus allegedly Graptophyllum pictum also have antidiabetic activity and it compound had observed. Antidiabetic activity of Graptophyllum pictum done by determining the magnitude of the inhibitory Graptophyllum pictum extract against α-glucosidase enzyme, the color intensity was read at 400 nm of wavelength with a microplate reader and calculated the percentage of Graptophyllum pictum extracts inhibition against α-glucosidase enzyme. Graptophyllum pictum maceration is done by using four kinds of solvents, ethanol 96, 70, and 30% and aquadest. The results showed the value of α-glucosidase inhibition by the ethanol extract of 96, 70, 30 and water respectively 61.09%, 66.11%, 58.55% and 40.13%. Phytochemical analysis showed that the extract with ethanol 96% was abled to extract the alcaloids, flavonoids, tannins and steroids; ethanol 70% extract of alcaloids, flavonoids, tannins, saponins and steroids; ethanol 30% extract alkaloids, flavonoids and saponins, and aquadest extract the alkaloids and flavonoids. Ethanol extract of 96% and 70% are the two extracts with α-glucosidase inhibition percentage above 50%, amounting to 61.09% and 66.11% so that it can be said that both the extract as an antidiabetic potential. Solvents which can extract all the expected compound is 70% ethanol.en
dc.description.abstractDaun wungu (Graptophyllum pictum (L.) Griff) merupakan salah satu tanaman dari famili Acanthaceae. Tanaman lain yang berada dalam satu famili dilaporkan memiliki aktivitas antidiabetes melalui mekanisme inhibisi enzim α- glukosidase, sehingga diduga daun wungu juga memiliki aktivitas antidiabetes tersebut dan diamati kandungan fitokimianya. Aktivitas antidiabetes daun wungu dilakukan dengan menentukan besarnya daya hambat ekstrak daun wungu terhadap enzim α-glukosidase, intensitas warna yang dihasilkan dibaca serapannya pada panjang gelombang 400 nm dengan microplate reader dan dihitung persentase daya hambat ekstrak daun wungu terhadap enzim α- glukosidase. Maserasi daun wungu dilakukan dengan menggunakan empat jenis pelarut, yaitu etanol 96, 70, dan 30% serta air. Hasil yang diperoleh menunjukkan nilai inhibisi α-glukosidase oleh ekstrak etanol 96, 70, 30 dan air berturut-turut 61.09%, 66.11%, 58.55% dan 40.13%. Analisis fitokimia menunjukkan bahwa ekstrak dengan pelarut etanol 96% dapat mengekstraksi senyawa alkaloid, flavonoid, tanin dan steroid; pelarut etanol 70% mengekstraksi alkaloid, flavonoid, tanin, saponin dan steroid; pelarut etanol 30% mengekstraksi alkaloid, flavonoid dan saponin; dan pelarut air mengekstraksi alkaloid dan flavonoid. Ekstrak etanol 96% dan 70% merupakan dua ekstrak dengan persentase inhibisi α- glukosidase diatas 50%, yaitu sebesar 61.09% dan 66.11% sehingga dapat dikatakan bahwa kedua ekstrak tersebut berpotensi sebagai antidiabetes. Pelarut yang dapat mengekstraksi seluruh senyawa yang diharapkan adalah etanol 70%.
