Inoculum Production of Arbuscular Mycorrhizal Fungi (AMF) using Aeroponics Culture and Solid State Media.

dc.description.abstractThe aim of this research was to study the technique of AMF inoculum production using aeroponics and solid state media. The experiments were carried out in the green house and laboratory. The experiments were conducted in two kinds of experimental design, namely Completely Randomized Design for experiment I and Split-blok Randomized Design for experiment II with five replications for each treatment. Host plants used were Ipomoea reptana, Centrosema pubescens, and Allium cepa. The first experiment was called inoculation phase. This experiment had two kinds of treatment, with and without AMF inoculation. Inoculation phase was grown in 200 grams of sterilize zeolite. The plants were grown until 12 weeks. The second experiment was called inoculum production. This experiment used three kinds of growing medium, they were aeroponics culture, compost and zeolite media. The plants were harvested at 12 and 18 weeks after planting. The AMF growth in inoculations phase experiment produced colonization percentage more than 70%. C. pubescens grew better in the AMF treatment. Experiment II showed that the best growths of I. reptana and C. pubescens associated with AMF were in aeroponics culture, while A. cepa was in zeolite media. Aeroponics culture significantly increased the total root length per pot of I. reptana. The length was 19,13 m. The best percentage colonization was observed at C. pubescens in aeroponics culture. The percentage colonization was 90,8%. The quality test of resulting AMF inoculum conducted that the inoculum could colonization the root to about 25-53%. The aeroponic culture produced the best quality of AMF inoculum followed by zeolite and compos media.en
dc.description.abstractPenelitian ini bertujuan untuk mempelajari teknik produksi CMA dengan menggunakan media alternatif kultur aeroponik dan media padat. Penelitian dilakukan di rumah kaca dan laboratorium. Penelitian ini menggunakan dua macam rancangan percobaan yaitu RAL untuk percobaan I dan RAK Split Blok untuk percobaan II dengan 5 ulangan untuk setiap perlakuan. Tanaman inang yang digunakan ialah Ipomoea reptana, Centrosema pubescens, dan Allium cepa. Percobaan I terdiri dari 2 perlakuan yaitu inokulasi dan tanpa inokulasi CMA. Fase inokulasi dilakukan dengan metode kultur pot menggunakan media zeolit steril sebanyak 200 gram dengan pertumbuhan tanaman selama 12 minggu. Percobaan II atau fase produksi dilakukan dengan cara memindahkan tanaman inang yang telah terkolonisasi CMA pada fase inokulasi ke dalam tiga media tumbuh yaitu kultur aeroponik, media kompos dan zeolit. Pemanenan dilakukan pada minggu ke 12 dan 18 setelah tanam. Percobaan I menunjukkan persentase kolonisasi pada setiap tanaman inang lebih dari 70%. Tanaman inang C. pubescens tumbuh baik pada perlakuan CMA dibandingkan dengan perlakuan tanpa inokulasi. Percobaan II menunjukkan pertumbuhan terbaik I. reptana dan C. pubescens yang bersimbiosis dengan CMA ialah kultur aeroponik, sedangkan A. cepa ialah media zeolit. Kultur aeroponik secara nyata dapat meningkatkan total panjang akar per pot tanaman I. reptana sebesar 19,13 m. Persen kolonisasi CMA terbaik diperoleh pada tanaman C. pubescens dengan media kultur aeroponik yaitu 90,8%. Uji lanjut inokulum CMA menghasilkan persentase kolonisasi sebesar 25-53%. Kultur aeroponik menghasilkan kualitas inokulum CMA terbaik diikuti oleh media zeolit dan kompos.
