dc.description.abstractPhytopathogenic bacterium, Xanthomonas campestris is an aerobic bacterium that caused rot disease on cruciferous plants, such as peppers, broccoli, and cabbage. This bacterium produced enzymes and extracellular polysaccharide biopolymers that can be used in food processing. The applications of xanthan gum as one of the extracellular polysaccharides of X. campestris in industry used as viscosifying and thickening agents, stabilizing, and suspension agents. Xanthan gum can be synthesized by transglycosylation reaction. The objective of this research was to know the capability of X. campestris in synthesizing transfer product on transglycosyzation reaction by using CGTase from various carbohydrate sources. Transfer activity was tested using thin layer chromatography (TLC) and high performance liquid chromatography refraction index (HPLC RI). Corn showed as the best substrate in the formation of transfer product compared than the other substrates. Using xylose acceptor was obtained that the level content of xanthan gum was 0.47% of 0.6% xanthan gum standard (Sigma®).en
