dc.description.abstractPenyakit bilur kacang tanah yang disebabkan oleh peanut stripe virus (PStV) merupakan salah satu penyakit penting pada pertamman kacang tanah. Penggunaan varietas tanah merupakan alternatif yang dianggap paling efektif untuk mengatasi penyakit tersebut. Masdahnya adalah, kultivar tahan belum tersda clan upaya pemuliaan menghadapi masalah ti& tersedianya gen resisten pada varietas budidaya. Rekayasa genetika merupakan metode efektif untuk mengatasi masalah tersebut Banyak bukti menunjukkan, tanaman yang tahan terhadap suatu virus dapat diperoleh dengan cara mengintegrasikan gen dari virus tersebut ke genom tanaman. Penelitian ini bertujuan (1) mengevaluasi respons tanaman kacang tanah transgenik yang membawa gen cp PStV terhadap infeksi PStV, (2) menguji stabilitas sifat resistensi terhachp PStV dari satu generasi ke generasi selanjutnya dan mengidentifikasi galur tanaman tmmgenik yang resisten PStV, (3) mengevduasi korelasi identitas transgen dan jumlah integrasi transgen cp PStV dengan resistensi tamman kacang tanah transgenik terhadap PStV, (4) mempelajari pola pewarisan sifat ketahanan terhadap PStV pada tanaman kacang tanah transgenik yang membawa gen cp PStV, dan (5) menguji kemungkinan penggabungan sifat resistensi terhadap PStV hasil rekayasa genetika dengan katakter daya hasil tinggi dan resistensi terhadap penyakit bercak daun Cercospora yang dikendalikan oleh gen alami dari tanaman kacang tanah nontransgenik.id
dc.description.abstractPeanut stripe disease caused by peanut stripe virus (PStV) is one of the major obstacles in peanut production. The use of resistant peanut cultivars is considered the most effective way to cope with this problem. However, resistant peanut cultivars have not been available and breeding for PStV resistance has been hampered by lack of the resistance gene in peanut gene pool. Plant genetic engineering could be a solution to the problem. A body of evidence is accumulating that plants resistant to a particular virus can be obtained by transforming the plants with the gene derived from the virus. This research aimed to (1) investigate response of transgenic peanuts to PSTV infection, (2) test the stability of resistance to PSTV from generation to generation and to identify PStV-resistant lines (3) determine the identity of the transgene and its copy number and its correlation with resistance to PStV, (4) determine inheritance of resistance, and (5) test whether a cross between transgenic peanuts resistant to PStV and a non-transgenic peanuts that were resistant to leafspot disease and high-yielding produce plants that were resistant to PSTV and leaf spot and high-yielding.
