      Poliploidisasi Tanaman Ciplukan (Physalis peruviana) secara In Vitro

      Abstract
      Ciplukan (Physalis peruviana) merupakan tanaman potensial untuk dikembangkan karena memiliki rasa yang khas dengan kandungan nutrisi dan metabolit sekundernya yang tinggi. Namun, produksi ciplukan di Indonesia masih terbatas karena minimnya teknik budidaya dan kualitas ciplukan yang masih rendah. Oleh karena itu perlu adanya peningkatan kualitas ciplukan melalui poliploidisasi secara in vitro menggunakan kolkisin. Penelitian ini menggunakan rancangan acak lengkap faktor tunggal dengan lima taraf konsentrasi kolkisin (0, 50, 100, 150, dan 200 ppm) yang diaplikasikan pada biji dengan cara direndam selama 24 jam. Biji yang telah direndam dalam kolkisin dikulturkan dan diamati pada tahap perkecambahan, pertumbuhan, sitologi, serta aklimatisasi. Hasil penelitian ini menunjukkan bahwa pengaruh kolkisin dapat menurunkan persentase perkecambahan dan pertumbuhan awal tunas yang tidak selalu berbanding lurus dengan peningkatan konsentrasi kolkisin. Analisis flow cytometry dan pengamatan stomata menunjukkan terbentuknya tanaman miksoploid pada seluruh taraf perlakuan, dengan jumlah dan densitas stomata menurun seiring dengan peningkatan konsentrasi kolkisin. Pada penelitian ini konsentrasi kolkisin 150 ppm menghasilkan jumlah tanaman miksoploid tertinggi serta meningkatkan bobot buah, kadar Brix, diameter bunga, diameter batang, tebal daun, dan luas daun. Konsentrasi kolkisin 150 ppm menjadi konsentrasi paling potensial untuk menginduksi poliploidisasi P. peruviana pada level miksoploid.
       
      Golden berry (Physalis peruviana) is a plant with significant development potential due to its distinctive flavor and high content of nutrients and secondary metabolites. However, its production in Indonesia remains limited by a lack of cultivation techniques and generally low fruit quality. Therefore, improving quality through in vitro polyploidization using colchicine is necessary. This study employed a single-factor completely randomized design with five colchicine concentration levels (0, 50, 100, 150, and 200 ppm) applied to seeds via a 24-hour soaking method. Following treatment, the seeds were cultured and monitored through stages of germination, growth, cytological analysis, and acclimatization. The results indicated that colchicine reduced germination percentage and early shoot growth, though these effects did not always correlate linearly with increasing concentration. Flow cytometry and stomata observations revealed the formation of mixoploid plants across all treatment levels, with stomata number and density decreasing as colchicine concentration increased. A concentration of 150 ppm yielded the highest number of mixoploid plants and enhanced fruit weight, brix levels, flower diameter, stem diameter, leaf thickness, and leaf area. Consequently, 150 ppm was identified as the most promising concentration for inducing mixoploid-level polyploidization in P. peruviana.
