mengakibatkan overeksploitasi sehingga terjadi kelangkaan sumber benih. Selain
3 kali ulangan terdiri atas kontrol, salinitas 20 ppt cahaya 500 lux, salinitas 25 ppt
cahaya 500 lux, salinitas 30 ppt cahaya 500 lux, salinitas 35 ppt cahaya 500 lux,
salinitas 20 ppt cahaya 2000 lux, salinitas 25 ppt cahaya 2000 lux, salinitas 30 ppt
cahaya 2000 lux, dan salinitas 35 ppt cahaya 2000 lux. Rumput laut Gracilaria sp.
seragam, yaitu mengalami peningkatan mulai hari pertama, mencapai puncak
spora/sistokarp. Dengan demikian data kuantitatif tersebut selaras dengan
pengamatan perkembangan sel. Pada hari pertama spora rilis dan terjadi
pembelahan awal 2-4 sel, selanjutnya terjadi pembentukan multiseluler pada hari
ke-7, sel mulai berdiferensiasi secara spasial hari ke-14. Selanjutnya, hari ke-21
memperlambat diferensiasi jaringan pembentukan tunas apikal. Tingkat
mendukung peningkatan kinerja pertumbuhan spora Gracilaria sp.
Kata kunci: Cahaya, Gracilaria sp., salinitas, spora. Gracilaria sp. seaweed is commonly cultivated using vegetative techniques,
employing fragments or cuttings randomly spread over the pond bottom using a
loose-bottom method. The source of seeds used generally comes from nature and
requires a large number of seeds, so the availability of quality seeds must be
sustainable. The continuous use of seeds from nature will result in overexploitation
and scarcity of seed sources. In addition to using natural seeds, tissue culture
techniques are an effort to provide quality seeds to date. However, this technique
usually requires higher production and operational costs, culture media, and
specialized labor and is vulnerable to contamination by microorganisms. These
limitations hinder meeting the need for seedlings in bulk and economically. One of
the efforts that can be made is to utilize the life cycle of Gracilaria sp. through
generative development, namely by spores. Spore culture creates genetic variability
where this technique is based on using fertile talus and releasing spores under the
right conditions or with fulfilled nutrient content. Environmental manipulation is
one of the strategies to improve the production performance of seaweed
cultivation, particularly for Gracilaria sp. spores. Therefore, this study aimed to
investigate the effects of salinity and light on the growth performance of
Gracilaria sp. spores as an in vitro source of seedlings.
The laboratory experiment was conducted from December 2023 to February
2024 at the Tissue Culture Laboratory of Pangkajene and Island State Agricultural
Polytechnic, South Sulawesi. A completely randomized design (CRD) consisting
of 9 treatments and 3 replicates. The treatments included a control, salinity of 20
ppt with 500 lux light intensity, salinity of 25 ppt with 500 lux, salinity of 30 ppt
with 500 lux, salinity of 35 ppt with 500 lux, salinity of 20 ppt with 2000 lux,
salinity of 25 ppt with 2000 lux, salinity of 30 ppt 2000 lux and salinity of 35 ppt
with 2000 lux. Gracilaria sp. seaweed used was talus containing adult carpospore
spores cultivated in Dusun Biring Kassi, Bulu Cindea Village, Kec. Bungoro, Kab.
Pangkep, South Sulawesi. The test parameters measured during the study included
the calculation of the number of released spores, the speed of spores attached to
the substrate, the number of spores attached, development of spore cells, and the
survival rate.
The research results indicate that the application of salinity and light
intensity exerts a significant effect on all spore performance variables (p < 0.05).
Spore release across all treatments exhibits a uniform temporal fluctuation pattern,
characterized by an increase starting on the first day, reaching its peak secretion
on the third day, and subsequently declining drastically on the fourth and fifth
days. The highest quantity of spore release is achieved under the 35 ppt salinity
and 2000 lux light intensity treatment, yielding an average of 918.33
spores/cystocarp/day and a total accumulation of 3,018.32 spores/cystocarp. This
elevated release under extreme conditions is indicated as a cellular osmoregulatory
response to mitigate environmental stress.
However, this massive release potential is non-linear with substrate fixation
efficiency due to partial cellular dehydration.
Significantly expedited attachment performance is concentrated under
balanced physiological conditions, specifically the 30 ppt salinity treatment
combined with 500 lux light intensity. Under these parameters, spores initiate
partial attachment in only 1.7 days and achieve complete adherence by day 3. The
S30C500 treatment also demonstrates the highest attached spore density on the
third day, reaching 865 spores/cystocarp. Consequently, these quantitative data
align with observations of cellular development. On the first day, spores are
released and undergo initial 2–4 cell divisions, followed by multicellular
formation by day 7, and spatial cell differentiation by day 14. Subsequently, on
day 21, the discoid structure thickens and compacts; at this stage, the apical-basal
polarity of the cells becomes clearly visible. The central region exhibits
accelerated cell proliferation, which triggers the formation of vertical upright
shoots, while the basal region maintains the discoid structure for anchorage. By
day 30, the young thallus stage successfully forms rhizoid-like structures.
Conversely, at a salinity of 20 ppt, cells swell and frequently undergo lysis due to
excessive turgor pressure, whereas a light intensity of 2000 lux induces
photosynthetic inhibition, which retards tissue differentiation and apical shoot
formation. The highest plantlet survival rate is consistently achieved by the
S30C500 treatment, with values ranging from 67% to 87%.
The results of this study indicate that the use of optimal salinity and light
conditions can enhance the growth performance of Gracilaria sp. spores,
including their survival rate and cellular development, thereby contributing to
increased seedling production. A salinity of 30 ppt combined with a light intensity
of 500 lux was identified as the optimal treatment for promoting the growth
performance of Gracilaria sp, spores.
