      Abstract
      Spirogyra sp. merupakan mikroalga yang dapat dimanfaatkan sebagai bahan baku bagi kebutuhan pangan dan energi. Penumbuhan biomassa yang terkontrol diperlukan guna mendapatkan produktivitas dan performa sel yang optimal. Spirogyra sp. memiliki kloroplas yang menempel pada dinding sel dan tersusun melingkar membentuk spiral yang umumnya bereproduksi secara vegetatif pada kondisi lingkungan yang optimal, juga dengan cara lain melalui konjugasi. Oleh karena itu, penelitian ini bertujuan untuk membandingkan biomassa dan performa sel Spirogyra sp. pada teknik kultur yang berbeda. Penelitian dilakukan pada bulan Januari hingga April 2026 dengan dua perlakuan teknik kultur yaitu batch dan semicontinuous. Parameter yang dianalisis yaitu kualitas air (suhu, pH, amonia, nitrat, dan ortofosfat) dan performa sel Spirogyra sp. (warna, kerapatan kloroplas, diameter sel, panjang sel). Hasil penelitian ini menunjukkan bahwa perlakuan batch memiliki akumulasi biomassa lebih tinggi serta performa sel yang lebih baik dibandingkan perlakuan semi-continuous. Konsentrasi amonia, nitrat, dan ortofosfat menurun pada kedua perlakuan selama periode kultur. Penelitian ini menunjukkan bahwa teknik kultur batch lebih efektif dalam mendukung produktivitas biomassa dan mempertahankan performa sel Spirogyra sp.
       
      Spirogyra sp. is a microalga that can be utilized as a raw material for food and energy needs. Controlled biomass growth is necessary to achieve optimal cell productivity and performance. Spirogyra sp. has chloroplasts attached to the cell wall and arranged in a circular spiral it generally reproduces vegetatively under optimal environmental conditions, as well as through conjugation. Therefore, this study aims to compare the biomass and cellular performance of Spirogyra sp. under different culture techniques. The study was conducted from January to April 2026 using two culture techniques batch and semi-continuous. The parameters analyzed included water quality (temperature, pH, ammonia, nitrate, and orthophosphate) and Spirogyra sp. cell performance (color, chloroplast density, cell diameter, and cell length). The results of this study indicate that the batch treatment yielded higher biomass accumulation and better cell performance compared to the semicontinuous treatment. The concentrations of ammonia, nitrate, and orthophosphate decreased in both treatments during the culture period. This study indicates that the batch culture technique is more effective in supporting biomass productivity and maintaining the performance of Spirogyra sp. cells.
