      Abstract
Carrageenan is a sulfated polysaccharide derived from red seaweed (Rhodophyceae), particularly the genera Kappaphycus and Eucheuma. It is recognized as a versatile hydrocolloid and essential raw material for pharmaceutical, cosmetic, and food applications. Carrageenan extraction can be achieved using hot water, alkaline extraction, enzyme-assisted extraction, and other emerging techniques, with the extraction yield depending on pretreatment, processing conditions, and seaweed characteristics. This study aimed to examine the effects of physical pretreatments, including sonication, soaking, and abrasion, and extraction conditions comprising temperature and time, on the yield and quality characteristics of carrageenan from Kappaphycus alvarezii cultivated in Zanzibar. The study employed physical pretreatments to the seaweed sample for 20 min, and solvent extraction using (8%) KOH solution at 70°C for 30 min, followed by distilled water extraction at 80 and 90°C for 30 and 60 min. The quality of the extracted carrageenan was evaluated based on physicochemical characteristics. The Factorial Completely Randomized Design (FCRD) and Duncan’s Multiple Range Test (DMRT) were used to determine significant effects of pretreatment and extraction conditions. The results show that pretreatments, temperature, and time significantly influence the yield and quality of carrageenan, although there is no significant interaction effect between them. Sonication pretreatment yielded the highest results (20.03%) at 80°C for 30 min and (26.30%) at 90°C for 60 min, while maintaining standard quality, gel strength (685.29 - 780.92 g/cm²), and viscosity (44.55 - 63.67 cP). Soaking maintains a high gel strength (795.60 - 867.81 g/cm²) and a low ash content (17.08 - 18.25%), while abrasion maintains a competitive whiteness index (75.49 - 81.02%). Also, control treatments resulted in lower yields (13.67 - 19.47%) but higher viscosity (50.78 - 63.60 cP). Therefore, pretreatment prior to the extraction significantly influences carrageenan yield and quality and should be considered in extraction processes. Further studies should optimize pretreatment duration to maximize the benefits of this approach.
