      Perbanyakan Tunas Jeruk Siam Berastagi (Citrus reticulata Blanco) Menggunakan Kinetin dan BAP secara In Vitro

      Abstract
      Perbanyakan jeruk siam Berastagi (Citrus reticulata Blanco) secara konvensional dinilai kurang efisien karena prosesnya lambat dan rentan terhadap penyakit, sehingga diperlukan teknik kultur in vitro sebagai solusi. Penelitian ini terdiri atas dua percobaan perbanyakan tunas, yaitu dengan eksplan tunas apikal dan eksplan tunas aksilar, yang bertujuan mempelajari pengaruh kombinasi konsentrasi kinetin dan BAP serta menentukan kombinasi optimal dalam perbanyakan tunas. Masing-masing percobaan menggunakan rancangan kelompok lengkap teracak satu faktor dengan empat taraf kombinasi konsentrasi BAP dan kinetin yang dikombinasikan dengan 0,5 mg L?¹ NAA. Pengamatan dilakukan selama 12 minggu setelah tanam. Hasil penelitian menunjukkan kombinasi konsentrasi BAP dan kinetin memengaruhi respons perbanyakan tunas jeruk siam Berastagi secara in vitro. Secara deskriptif, eksplan tunas aksilar cenderung menghasilkan jumlah tunas dan daun lebih banyak pada akhir pengamatan dibandingkan eksplan tunas apikal. Tidak diperoleh satu kombinasi konsentrasi BAP dan kinetin yang secara konsisten menghasilkan respons terbaik pada seluruh peubah pengamatan, sehingga belum dapat ditentukan kombinasi yang paling sesuai untuk perbanyakan tunas jeruk siam Berastagi secara in vitro.
       
      Conventional propagation of Berastagi Siamese orange (Citrus reticulata Blanco) is inefficient due to its slow process and high disease susceptibility, requiring in vitro culture as a solution. This study consisted of two shoot multiplication experiments using apical and axillary shoot explants to examine the effect of kinetin and BAP concentration combinations and determine the optimal combination for shoot multiplication. Each experiment used a single-factor randomized complete block design with four concentration combinations of BAP and kinetin supplemented with 0.5 mg L?¹ NAA, observed for 12 weeks after planting. The results showed that the combination of BAP and kinetin concentrations affected the shoot multiplication response of Berastagi siamese orange in vitro. Descriptively, axillary shoot explants tended to produce higher shoot and leaf numbers than apical shoot explants by the end of observation. No single combination consistently produced the best response across all variables, so the most suitable combination for in vitro shoot multiplication of Berastagi Siamese orange could not yet be determined.
