      Pengaruh Variasi Naungan Terhadap Kadar Total Fenolik dan Aktivitas Antioksidan Ekstrak Tanaman Miana (Coleus scutellarioides (L.) Benth)

      Abstract
      Tanaman miana (Coleus scutellarioides (L.) Benth) telah terbukti mengandung berbagai senyawa aktif, salah satunya adalah senyawa fenolik yang memiliki aktivitas antioksidan. Biosintesis senyawa tersebut berpotensi dipengaruhi oleh intensitas paparan cahaya yang diterima tanaman. Penelitian ini bertujuan mengukur kadar total fenolik dan aktivitas antioksidan dari tanaman miana yang diberi perlakuan naungan. Kadar total fenolik ditentukan dengan metode Folin-Ciocalteu dan aktivitas antioksidan ditentukan dengan metode penangkapan radikal (DPPH) dan aktivitas reduksi (FRAP). Hasil menunjukkan perbedaan signifikan antar perlakuan naungan. Kadar total fenolik dan aktivitas antioksidan tertinggi dimiliki oleh ekstrak perlakuan naungan 0%, yaitu sebesar 6,307±0,07 mg GAE/BK, 8,199±0,21 µmol TE/g BK untuk metode DPPH, dan 39,323±0,60 µmol TE/g BK untuk metode FRAP. Kadar total fenolik dan aktivitas antioksidan cenderung menurun pada naungan 25% dan 50%, tetapi kembali naik pada naungan 75%. Hal tersebut diduga disebabkan oleh respon adaptasi tanaman terhadap intensitas cahaya yang diterima. Analisis korelasi menunjukkan kadar total fenolik cenderung berhubungan linear dengan aktivitas antioksidan.
       
      Coleus scutellarioides (L.) Benth has been reported to contain various bioactive compounds, one of which is phenolic compounds that possess antioxidant activity. These compounds are potentially influenced by the intensity of light exposure received by the plant. This study aimed to determine the total phenolic content and antioxidant activity of the plant through shading treatments. Total phenolic content was determined using the Folin–Ciocalteu method, while antioxidant activity was evaluated using the radical scavenging method (DPPH) and ferric reducing antioxidant power (FRAP) assay. The results showed significant differences among treatments. The highest total phenolic content and antioxidant activity were observed in the 0% shading treatment extract, with values of 6,307 ± 0,07 mg GAE/g DW, 8,199 ± 0,21 µmol TE/g (DPPH), and 39,323 ± 0,60 µmol TE/g (FRAP). TPC and antioxidant activity tended to decrease under 25% and 50% shading treatments, but increased again under 75% shading. Correlation analysis showed that total phenolic content tended to have a linear relationship with antioxidant activity.
